法舒地尔对大鼠肺动脉平滑肌细胞收缩、迁移能力的影响 被引量:1

摘要 目的探讨法舒地尔对大鼠肺动脉平滑肌细胞(PASMCs)收缩、迁移能力的影响。方法以体外培养的PASMCs为研究对象,分为正常培养对照组和不同浓度(10μmol/L,30μmol/L,50μmol/L)法舒地尔处理组,分别采用胶原凝胶实验及Boyden Chamber实验方法对PASMCs的收缩和迁移能力进行分析。结果法舒地尔能够显著抑制PASMCs的收缩和迁移(P<0.01),并具有浓度依赖性。结论法舒地尔对PASMCs的收缩和迁移能力具有显著抑制作用,这可能是其治疗肺动脉高压的重要作用机制。 Objectives To investigate the effects of fasudil on the contractility and migration of rat pulmonary artery smooth mus-cle cells (PASMCs).Methods PASMCs were incubated or not (controls)with different concentrations(10μmol/L,30μmol/L,50μmol/L)of fasudil.The contractility and migration of PASMCs were investigated by collagen gel contraction assay and boyden cham-ber assay,respectively.Results Fasudil effectively inhibited the contractility and migration of PASMCs (P〈0.01),in a dose de-pendent manner.Conclusion Fasudil effectively inhibited the contractility and migration of PASMCs,which might serve as an impor-tant mechanism of its anti pulmonary arterial hypertension effects.
