摘要
目的:研究六味地黄丸对N-甲基-N-亚硝基脲(MNU)诱C57BL/Ks J-db/db小鼠胃癌的保护作用并对其机制进行初步探讨。方法:同批22只雄性糖尿病模型动物C57BL/Ks J-db/db小鼠随机分为两组:对照组及六味地黄丸组,隔周给予新鲜配制的MNU120ppm自由饮水20周诱导胃癌模型,21周起两组小鼠分别给予蒸馏水及六味地黄丸,持续10周,观察记录各组小鼠一般情况。末次给药后测量体质量,空腹血糖,分离胃组织,HE染色观察各实验组胃组织的病理学改变,Real-time PCR法检测胃癌相关炎症因子IL-6,IL-1β,IL-10,IL-18,TNF-α及IFN-γmRNA的相对表达量。结果:六味地黄丸组C57BL/Ks J-db/db小鼠胃黏膜异型增生占10%,无肿瘤形成;对照组小鼠异型增生占67%,1例(11%)胃肿瘤形成,差异有统计学意义(P<0.05),Realtime PCR结果显示六味地黄丸能显著降低糖尿病小鼠胃癌相关炎症因子1L-6,1L-1β,1L-10 mRNA的表达(P<0.05),TNF-α的相对表达量降低58%,IL-10及IFN-γ的表达所有所下调,但无统计学差异。结论:六味地黄丸可通过抑制胃癌相关炎症因子的表达降低糖尿病小鼠胃癌的发生风险。
Objective：To establish mouse model of gastric carcinoma and research the preventive effects of liuwei di- huang pills on gastric cancer related inflammatory cytokines gene expression and its potential mechanism during the tumor- igenesis of stomach. Methods ： 22 four - week - old male C57BL/KsJ - db/db mice were randomly divided into control group and liuwei dihuang pills group to induce gastric cancer modeld by giving N -Methyl -N -Nitrosourea （MNU） ev- ery other week for 20 weeks. One week after completion of MNU administration, mice were given distilled water and li- uwei dihuang pills dissolved in distilled water for 10 weeks. After that ; mice were euthanised using 10% pentobarbital so- dium. Final diagnoses were made after histopathological examination. Quantitative Real time PCR was performed using specific primer sets that amplify IL- 6,IL- 113, IL- 10, IL- 18 ,TNF -c~ and IFN -7 mRNA to assess its relative ex- pression ratio. Result： 10% mice in the liuwei dihuang pills group had chronic gastritis, while 67% in control group to- gether with one gastric cancer. Compared with control group, the liuwei dihuang pills group reached significant statistical on gastric tumorigenesis （P 〈 0.05）. Outcomes of quantitative Real time PCR showed that there is a significant difference between liuwei dihuang pills group and control group in the expression of IL- 6, IL- 113 and IL -10 （P 〈 0.05 ）, while the expression of TNF -ct was downregulated to 58%. Besides, there was no difference between these two group on the expression of IL - 18, and IFN - %, mRNA. Conclusion： liuwei dihuang pills may be useful in preventing of gastric tumor- igenesis in diabetic mice.
