摘要
目的检测Caspase-3及Survivin在急性胰腺炎大鼠胰腺组织中的表达并探讨其与胰腺组织损伤的关系。方法 60只SPF级雄性Wistar大鼠随机分为假手术组、胰腺炎6 h组、胰腺炎12 h组及胰腺炎24 h组。采用胰腺被膜下注射牛磺胆酸钠(Na-Tc)建立急性胰腺炎模型。分别检测4组大鼠血清淀粉酶(AMY)、胰腺病理损伤、胰腺组织中Caspase-3及Survivin的表达程度。结果假手术组、胰腺炎6 h、12 h、24 h组组间两两比较血清淀粉酶及胰腺组织病理评分胰腺组织Caspase-3及Survivin平均光密度表达差异均有统计学意义(P<0.05)。Caspase-3、Survivin、血清淀粉酶及胰腺病理评分均有相关性(P<0.05)。结论急性胰腺炎过程中伴随着Caspase-3及Survivin的表达紊乱,并且与胰腺损伤密切相关。
Objective To detect the expression levels of Caspase-3 and Survivin in rats with acute pancreatitis,and to explore their correlation with pancreatic tissue damage.Methods Sixty SPF male Wistar rats were randomly divided into four groups:sham-operation group ( n =15),6 hours after pancreatitis group (6h group, n =15),12 hours after pancreatitis group (12h group, n =15),24 hours after pancreatitis group (24h group, n =15).The animal models with pancreatitis were induced by injection of 5% Na-Tc under capsula pancreatis.The activity of serum amylase,pathological injury of pancreas, expression levels of Caspase-3 and Survivin were detected for all the groups.Results There were significant differences in the activity of serum amylase, pathological injury of pancreas and the expression levels of Caspase-3 and Survivin among sham-operation group,6h group,12h group,24h group with each other ( P 〈0.05).There were close correlations among Caspase-3,Survivin, serum amylase and pathological injury scoring.Conclusion The process of acute pancreatitis is accompanied with the expression disorder of Caspase-3 and Survivin, furthermore, which is closely correlated to pancreas injury.
