摘要
采用碱性蛋白酶水解鹿尾蛋白质制备鹿尾肽。应用响应面分析法选取温度、时间、p H和酶量4个主要因素,以水解率为响应值,对其工艺进行了优化。得出了鹿尾蛋白水解的最佳工艺条件为:温度53℃、时间5.9 h、p H8.8、酶量1 560 U·g-1。在此条件下鹿血的实际水解率为24.37%,酶解后多肽的分子量分布在1 000 Da左右。试验证明响应面法对鹿尾肽的制备条件的优化是可行的,得到的工艺参数具有实际应用价值。
The deer's tail protein peptide alcalase has been prepared by alcalase protease. Application of response surface methodology selected temperature, time and pH as main factors, the hydrolysis rate as response, the technology has been optimized. Optimum condition ofproteolysis of deer's tail obtained was that temperature 53 ℃, time 5.9 h, pH 8.8, enzyme 1 560 U.g-1. The deer's tail in the actual hydrolysis rate was 24.37%. After enzymolysis, the molecular weight ofpolypeptide was about 1 000 Da. The results showed that the response surface method of preparation conditions on deer's tail peptide optimization was feasible, and the process parameters had practical value.
