摘要
以黄芪多糖粗提物为样品,采用蒽酮-浓硫酸比色法测定其多糖含量,并利用聚酰胺柱和AB-8大孔树脂柱进行纯化,采用不同洗脱剂洗脱;用氯磺酸-吡啶法对黄芪多糖进行硫酸酯化;红外光谱分析黄芪多糖粗提物、纯化样及硫酸酯样品,研究黄芪多糖粗提物的纯化和硫酸化并对其红外光谱进行分析。结果表明,聚酰胺柱或AB-8大孔树脂柱对黄芪多糖进行纯化,使用去离子水或30%以下的乙醇作为洗脱剂均可取得较好的效果;氯磺酸-吡啶法可以对黄芪多糖进行硫酸酯化。
Using radix astragali polysaccharides as material, the contents of polysaccharides were determined by anthrone-sulfuric acid colorimetric method and purified using polyamide or AB-8 macro-porous resin column chromatography. The polysaccharides were eluted with deionization water and different concentration ethanol. Radix astragali polysaccharides were sulfated by chlorosulfonic acid pyridine method. The raw extract, purified and sulfated samples of radix astragali polysaccharides were all analyzed with infrared spectroscopy technology. Results showed that purification of radix astragali polysaccharides using these procedures mentioned above was feasible. Deionized water or 30% following ethanol as eluants had good effects. Chlorosulfonic acid-pyridine method can be used to sulfate radix astragali polysaccharides.
