摘要
目的探讨中药复方胃肠安对人SGC-7901胃癌细胞裸鼠移植瘤生长的抑制作用及抗肿瘤机制。方法将45只SGC-7901胃癌移植瘤裸鼠随机分为对照组,胃肠安低、中、高剂量组(剂量分别为15,30,60mg·kg-1,灌胃给药)。记录4组小鼠肿瘤生长以及生存情况,并采用实时荧光定量PCR(RF-PCR)分析肿瘤组织中凋亡相关基因(Caspase-3、Bcl-2和Bax)的表达情况,采用TUNEL染色法分析不同处理肿瘤组织中肿瘤细胞凋亡指数。结果与对照组比较,胃肠安低、中、高剂量组小鼠肿瘤生长明显被抑制、生存率明显增加(P<0.05),呈剂量依赖效应;小鼠肿瘤组织中Caspase-3和Bax mRNA表达水平以及细胞凋亡指数明显增加,而Bcl-2 mRNA表达水平明显下降(均P<0.05),随着药物浓度的增加呈剂量依赖性变化。结论中药复方胃肠安可显著提高肿瘤组织中肿瘤细胞凋亡基因表达,提高凋亡指数,并抑制肿瘤生长,延长荷瘤小鼠的生存时间。
Objective To investigate the anti-tumor effect and mechanism of Chinese herbal medicine compound Weichang＇an on nude mice with SGC-7901 transplanted gastric cancer.Methods A total of 45 nude mice with SGC-7901 transplanted gastric cancer were randomly divided into control group and low-,middle-,and high-dose Weichang＇an groups（treated with Weichang＇an at 15,30,60 mg·kg^-1 by gavage respectively）.Tumor growth and survival condition in the four groups were recorded.Apoptosis related genes（Caspase-3,Bcl-2 and Bax） in tumor tissues were analyzed by real-time fluorescent quantitation PCR,and tumor cell apoptosis was analyzed by TUNEL staining method.Results Compared with the control group,tumor growth in the mice of the three medication groups was significantly inhibited（P〈0.05） in dose-dependent manner,and the survival rate in the three medication groups was increased significantly（P〈0.05） also in dose-dependent manner（P〈0.05）.Compared with the control group,caspase-3 and Bax mRNA expression level and apoptosis index in tumor tissues were significantly increased whereas Bcl-2 mRNA expression level was decreased significantly in the three medication groups（P〈0.05）.With the gradual increase of medicine concentration,the changes of Caspase-3,Bcl-2 and Bax mRNA expression level and apoptosis index in tumor tissue were dose-dependent.Conclusion Chinese herbal medicine compound Weichang＇an can significantly promote the expression of apoptotic gene in tumor tissue,increase the apoptotic index,inhibit tumor growth and prolong the survival time in mice bearing SGC-7901 gastric cancer.This study will provide theoretical basis for the clinical treatment of gastric cancer.
