三种脾虚泄泻证模型大鼠消化系统功能改变的比较 被引量:22

Comparative study on the changes of digestive system functions in three kinds of rat models of spleen-hypofunction diarrhea syndrome

摘要 目的:运用番泻叶法、乙酸法和番泻叶加乙酸法分别制作三种大鼠中医脾虚泄泻证模型,比较所受损伤对消化及免疫系统功能等的影响,以期从中选择出与中医临床脾虚泄泻证较为吻合的动物模型来评价相关的治疗药物。方法:实验于2005-08在解放军军事医学科学院放射与辐射医学研究所药理毒理研究室完成。健康SD大鼠48只,随机分为对照组、番泻叶组、乙酸组和番泻叶加乙酸组,每组12只。番泻叶组灌服20%番泻叶浸剂4g/(kg·d),连续7dR乙酸组在实验第5天起禁食不禁水32h,由肛门(深入8cm)注入8%的乙酸0.5mL,捏紧大鼠肛门,倒提20s。番泻叶加乙酸组同番泻叶组灌服番泻叶浸剂7dR并在实验第5天注入8%的乙酸,余同乙酸组。对照组灌服、注入均为生理盐水,操作同番泻叶加乙酸组。观察各组大鼠一般症状,测定稀便率、体质量、脾脏指数、胸腺指数、血清D-木糖含量、血清淀粉酶活性、血清琥珀酸脱氢酶比活性等指标。结果:实验大鼠48只均进入结果分析。①三种大鼠脾虚泄泻证模型均出现腹泻、体质量下降、畏寒、倦怠等类似临床脾虚泄泻证一般症状的表现。②各组大鼠造模后体质量:对照组大鼠造模后体质量与造模前比较有显著增长;三个模型组造模后明显低于对照组[对照组(202.99±13.15)g,番泻叶组(143.84±5.66)g,乙酸组(157.07±11.83)g,番泻叶加乙酸组(127.64±14.32)g,P<0.01]。③各组大鼠胸腺质量、胸腺指数、脾质量、脾脏指数变化:各模型组胸腺质量和胸腺指数变化与对照组比较均有显著差异,脾质量和脾脏指数变化与对照组相比,只有番泻叶加乙酸组两者差异都有显著性[胸腺指数:对照组(0.0021±0.0005)、番泻叶组(0.0010±0.0002)、乙酸组(0.0013±0.0006)和番泻叶加乙酸组(0.0006±0.0003);脾脏指数:番泻叶加乙酸组(0.0013±0.0007),P<0.01]。④各组大鼠肠道病理:肉眼观察和病理切片均可见 AIM: To select a spleen-hypofunction diarrhea syndrome model used for appraising the effective drug for this syndrome by compared the changes of digestive system functions after injury in three rat models of spleenhypofunction diarrhea syndrome, which were established'by Folium Sennae, acetic acid and Folium Sennae and acetic acid, respectively. METHODS: The experiment was conducted in the Laboratory of Pharmacology and Toxicology, Institute of Radiation Medicine, Academy of Military Medical Sciences in August 2005. Forty-eight healthy SD rats were selected and randomly divided into control group, Folium Sennae group, acetic acid group and Folium Sennae plus acetic acid group with 12 rats in each group. The Folium Sennae group was infused with 20% Folium Sennae 4 g/kg per day for 7 days; the acetic acid group fasted from the 4^th day for 32 hours, and 8% acetic acid 0.5 mL was infused into the anus (8 cm), and the rats were lifted inversely for 20 seconds after clenched the anus. The Folium Sennae plus acetic acid group was infused the matching Folium Sennae for 7 days and 8% acetic acid at day 5, following by the procedure as the acetic acid group. The control group was infused and injected with normal saline with the same procedure as the Folium Sennae plus acetic acid group. The general physical symptoms were observed and loose stools rate, body mass, spleen index, thoracic gland index, serum Dxylose content, serum amylase activity, and serum succinate dehydrogenase specific activity of each group were detected. RESULTS: All the 48 rats were involved in the result analysis. ①General physical symptoms of spleen-hypofunction diarrhea syndrome such as diarrhea, decreasing of body mass, chilly and tired appeared in the rat model of spleen-hypofunction diarrhea syndrome. ② Body mass after modeling: The body mass of the control group were increased after modeling compared with that before modeling; but the body mass of the three model groups after modeling were lower than the control group [cont
