摘要
目的:探究细胞因子信号转导抑制因子3(SOCS-3)在鼠巨细胞病毒(MCMV)感染新生小鼠肝组织中的作用。方法:将48只小鼠随机分为对照组与病毒组(MCMV组),予MCMV组腹腔注射MCMV 20μl,3、7和14天时取标本(各8只),观察其生长发育及肝脏病理变化;ELISA法检测血清丙氨酸转氨酶(ALT),RT-PCR法和免疫组化法测肝组织的SOCS-3,并对ALT与SOCS-3行相关分析。结果:MCMV组小鼠生长发育迟缓,肝脏病变明显;ALT表达量和SOCS-3表达量明显高于对照组,差异有统计学意义(P<0.05),SOCS-3与ALT在3和7天呈正相关(P<0.05)。结论:MCMV感染可诱导SOCS-3表达增多,且在3和7天与肝脏损伤程度呈正相关。
Objective: To explore the role of suppressor of cytokine signaling-3( SOCS-3) in hepatic tissue of neonatal rats with murine cytomegalovirus( MCMV) infection. Methods: Forty-eight mice were randomly divided into control group and virus group( MCMV group),the mice in MCMV group were treated with transabdominal injection of 20 μl MCMV,growth and development,hepatic pathological change of the mice were observed on the third day,the seventh day,and the fourteenth day,respectively; ELISA was used to detect the levels of serum alanine aminotransferase( ALT); RT-PCR and immunohistochemical staining were used to detect SOCS-3 levels in hepatic tissue; the correlation between ALT and SOCS-3 was analyzed. Results: Growth and development of mice in MCMV group was slow,hepatic lesions were obvious; the expression levels of ALT and SOCS-3 in MCMV group were statistically significantly higher than those in control group( P〈0. 05),SOCS-3 was positively correlated with ALT on the third day and the seventh day( P〈0. 05). Conclusion: MCMV infection can increase SOCS-3 expression,which is positively correlated with degree of hepatic injury on the third day and the seventh day.
