摘要
本研究以红芽芋球茎的顶芽或侧芽为外植体,采用MS培养基,附加不同的植物激素,进行组织培养试验,采用正交设计对影响红芽芋组织培养的丛生芽诱导进行优化,并建立红芽芋离体再生体系。研究结果表明:初代培养过程中MS+6-BA 1.5~2.0 mg/L+NAA 0.05 mg/L为丛生芽诱导最佳培养基;MS+6-BA2.0 mg/L+NAA 0.05 mg/L为理想的继代增殖培养基;1/2MS+IBA 0.1~0.3 mg/L是最佳的生根培养基适于诱导生根获得再生植株。
In the present research, the optimization of multiple shoot induction in Clocasia escalenta Schott was conducted by application of orthogonal design with MS media by adding different plant hormones using terminal bud or lateral bud of Clocasia escalenta Schott as explants. Finally, the in vitro regeneration system of Clocasia escalenta Schott was established. The result indicated that optimal formula of medium for multiple shoot induction, subculture multiplication and rooting media for Clocasia escalenta Schott tissue culture were MS+6-BA1.5~2.0 mg/L+NAA 0.05 mg/L, MS+6-BA 2.0 mg/L+NAA 0.05 mg/L, 1/2MS+IBA 0.1~0.3 mg/L respectively.
