摘要
目的:基于慢病毒载体介导的RNAi在体阻断SOCS1、PIAS1、PTPN22在RA动物模型的表达,研究艾灸对实验性RA家兔关节滑膜液EGF、FGF含量的影响及其SOCS1、PIAS1、PTPN22调控机制。方法:日本大耳白兔36只,随机分为空白对照组、RA模型组、艾灸治疗组、艾灸+SOCS1基因干扰组、艾灸+PTPN22基因干扰组、艾灸+PIAS1基因干扰组,6只动物/组。按0.5 m L/kg体质量剂量,将福氏完全佐剂平均注入模型组、艾灸各组动物的双后膝关节腔内(对照组动物同法注入无菌生理盐水作为对照),运用慢病毒介导的SOCS1、PTPN22、PIAS1 RNAi作为干预手段,麦粒灸"肾俞"、"足三里"穴治疗实验性RA(5壮/(穴·天),6 d1个疗程,共治疗3个疗程,疗程之间休息1 d)。ELISA法测定艾灸对关节滑膜液EGF、FGF含量的影响。结果:与对照组比较,模型组EGF、FGF含量显著升高(P<0.01);与模型组比较,艾灸组EGF、FGF含量显著降低(P<0.01);SOCS1干扰组、PTPN22干扰组、PIAS1干扰组与艾灸组比较,下调EGF、FGF含量效果不及艾灸组(P<0.05)。结论:艾灸"肾俞"、"足三里"穴可显著降低实验性RA家兔滑膜液相关细胞因子EGF、FGF的含量,但在SOCS1、PTPN22、PIAS1被干扰的情况下其作用显著降低,提示SOCS1、PTPN22、PIAS1对JAK-STAT信号通路的负反馈调控可能与艾灸抑制实验性RA滑膜细胞的异常分泌功能密切关联。
Objective: To study the effects of moxibustion on experimental RA synovial cell secretory function and SOCS1,PIAS1,PTPN22 regulatory mechanism by lentiviral vector-mediated RNAi in vivo blocking SOCS1,PIAS1,PTPN22 expression in RA animal model. Methods: 36 Japanese white rabbits,which divided into six group-normal group,model group,moxibustion group,SOCS1 RNAi group,PTPN22 RNAi group and PIAS1 RNAi group. Each group has 6rabbits. Just injectd the Freund's Complete Adjuvant( FCA) into the joint cavity of two hind legs of model group and all moxibustion groups by 0. 5 mL / kg. Using lentiviral-mediated SOCS1,PTPN22,PIAS1 RNAi as means of intervention,Apply treatment of Ai tablets moxibustion on "shenshu","Zusanli"to treat experimental RA( 5 Zhuang / point / day,6 days a course of treatment,three courses,one day rest between treatments). Detected EGF,FGF content of synovial fluid with Elisa assay. Results: test on cytokine levels of Synovial fluid showed that: Compared with the normal group,EGF,FGF levels of model group were significantly higher( P〈0. 01); compared with the model group,EGF,FGF levels of moxibustion group were significantly lower( P〈0. 01); compared with the moxibustion group,the content of EGF,FGF with PTPN22 RNAi group,PIAS1 RNAi group and SOCS1 RNAi group reduced is less than the effect of moxibustion group( P〈0. 05). Conclusions: moxibustion on "shenshu","Zusanli"can significantly reduced the content levels of inflammatory cytokines EGF,FGF in synovial fluid of experimental RA rabbits. But in the condition of SOCS1,PTPN22,PIAS1 RNAi,its anti-inflammatory effect was significantly reduced,which suggested that negative feedback regulation of SOCS1,PTPN22,PIAS1 in JAK-STAT signaling pathway may be an important mechanism of inhibition of cell secretory function by moxibustion in RA synovial.
