摘要
目的分析精神分裂症患者的亚甲基四氢叶酸还原酶(MTHFR)C677T基因多态性的流行病学特征及其在不同人群中的变异性。方法检索中国期刊全文数据库、万方数据库以及西文数据库包括:Pub Med、Embase、Schizophrenia Gene和Hu GENet等关于MTHFR C677 T基因多态性与精神分裂症遗传关联性的病例对照试验的中英文文献,检索时间至2013年8月30日,应用STATA SE12.0版本软件进行以上相关统计学分析。以Meta分析法合并该位点等位基因频率,分析C677 T等位基因在精神分裂症患者中的易感性。结果共纳入29篇合格文献(病例组8 259,对照组9 936例),各研究间有明显异质性。Meta分析显示:在全部人群及其三个亚组内,病例组合并的T等位基因频率和TT基因型频率均高于对照组,但由于95%CI相互重叠,差异均无统计学意义。三个亚组间比较:病例组中国人群合并T等位基因频率和TT基因型频率为最高(52%和28%),白种人群最低(33%和11%);健康对照组中国人群合并T等位基因频率和TT基因型频率为最高(52%和28%),白种人群最低(33%和11%)。病例组间比较,中国亚组的T等位基因频率和TT基因型频率高于白种人亚组,差异均有统计学意义。而三个亚组的对照组间的基因多态性分布差异均无统计学意义。结论受研究间异质性影响,目前尚不能认为精神分裂症人群MTHFR C677T的T等位基因及TT基因型频率明显高于正常人群,但不同地域和种族间基因多态性分布差异有统计学意义。
【Objective】To research on the polymorphisms and different population variability of methylenetetrahydrofolate reductase(MTHFR) gene C677 T. 【Methods】Chinese and English literatures about MTHFR C677 T gene polymorphism and schizophrenia genetic association controlled trials were retrieved in China Journal Full-text Databae(CJFD), Wanfang Data and Western databases including Pub Med, Embase, Schizophrenia Gene and Hu GENet,the relevant statistical analysis retrieval time was by Aug 30, 2013, software STATA SE12.0 version was applied for statistical analysis. Meta-analysis method was used to merge the frequency and analyze the susceptibility of C677 T allele in schizophrenia patients. 【Results】A total of 29 qualified literatures(8,259 cases in case groups, 9,936 cases in control groups) were brought into this investigation with significant heterogeneity between the studies. Metaanalysis showed that in the whole population and its three sub-groups, the combined T allele frequency and the TT genotype frequency in the case groups were higher than those in the control groups, but the differences had no statistical significance because the 95% CI were overlapping. Comparison between the three subgroups revealed that both in the case groups and the healthy control groups, the combined T allele frequency and the genotype TT frequency were the highest in Chinese populations(52% and 28%) and the lowest in Caucasian populations(33% and11%). The comparison between the case groups showed that the T allele frequency and the TT genotype frequency in Chinese subgroups were higher than those in Caucasian subgroups with statistically significant differences. The genetic polymorphism distribution had no statistical differences between the control groups and the three subgroups.【Conclusions】Affected by the heterogeneity between the studies, it cannot be concluded that MTHFR C677 T T al lele frequency and TT genotype frequency of schizophrenia crowd are significantly higher than those in normal peopl
