摘要 目的 :研究可溶性糖化终末产物 (AGE)受体 (sRAGE)对AGE诱导牛主动脉内皮细胞 (BAECs)NF κB激活的抑制作用。方法 :分别用AGE修饰白蛋白和羧甲基赖氨酸 (CML)修饰白蛋白培养BAECs ,凝胶迁移率改变实验检测加入sRAGE或RAGE反义寡核苷酸前后NF κB的DNA结合活性 ,并定量分析蛋白条带的密度积分。结果 :AGE修饰白蛋白和CML修饰白蛋白能通过结合BAECs表面RAGE诱导NF κB的激活 ,sRAGE和RAGE反义寡核苷酸均能有效的抑制AGE对NF κB的激活 ,sRAGE几乎能完全抑制AGE对NF κB的激活 ,而RAGE反义寡核苷酸抑制NF κB激活的作用是有限的。结论 :RAGE在AGE诱导BAECsNF κB的激活中起关键作用 。 Objective: To study inhibition of soluble RAGE(sRAGE)on advanced glycation end products(AGE)induced nuclear factor(NF-κB)activation in bovine aortic endothelial cell(BAECs).Method: We incubated BAECs with AGE-albumin and CML-albumin respectively.NF-κB binding activity was monitored by EMSA in the absence or presence of sRAGE or antisense-RAGE.Signal obtained in EMSA were analysised by densitometric quantification.Results: In this study,AGE-albumin and CML-albumin induced NF-κB activation by binding RAGE on surface of BAECs.Inhibition of NF-κB activation was accomplished using sRAGE or antisense-RAGE.sRAGE blocked all cell surface interaction of AGE/RAGE,while the effect of antisense-RAGE was only partial.Conclusion: AGE induced NF-κB activation in BAECs is dependent on RAGE.The blockade of RAGE by sRAGE provides a novel target for a therapeutic intervention in patients with complications of diabetes.
