摘要
目的:研究CD31基因的单核苷酸多态性和体内血清含量与原发性肝癌的关联性。方法选取160例肝癌患者和160例健康体检者作为观察对象,采用PCR-RFLP技术对CD31基因Leu125Val、Asn563Ser和Gly670Arg单核苷酸多态性进行基因分型,并且采用酶联免疫吸附试验检测血清CD31含量。结果肝癌患者和健康体检者的CD31基因Asn563Ser和Gly670Arg多态性差异不明显(χ2=0.387,P>0.05),而Leu125Val 多态性差异显著(χ2=10.875,P<0.05)。Val等位基因携带者患肝癌的风险是Leu等位基因的1.457倍(OR=1.457,95%CI:1.187~2.186,P=0.001);携带Val等位基因的原发性肝癌患者的血清CD31含量比没有携带的患者要高很多(χ2=10.534,P<0.05)。结合基因型分析表明,CD31基因Leu125Val、Asn563Ser和Gly670Arg单核苷酸多态性具有明显的连锁不平衡,同健康体检者相比, Val-Ser-Arg单倍型携带者罹患肝癌的风险较高(OR=1.487,95%CI:1.092-2.038,P=0.011)。结论 CD31基因的Leu125Val多态性及Val-Ser-Arg单倍型与原发性肝癌具有一定的关联,其中Val等位基因有极大的可能是原发性肝癌的遗传易感基因,携带Val基因型的患者可能会由于CD31基因在体内高度表达而增加罹患肝癌的可能性。
Objective To investigate the association of single nucleotide polymorphisms ( SNPs) of CD31 and serum level with hepatocellular carcinoma(HCC).Methods Three single nucleotide polymorphisms Leu125Val,Asn563Ser and Gly670Arg of CD31 were analyzed using polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP ) , and the serum level of CD31 was determined by enzyme-linked immunosorbent assay ( ELISA) in 160 HCC patients and 160 healthy patients .Re-sults The CD31 gene Asn563Ser and Gly670Arg polymorphisms had no significant difference between the HCC group and the control group(χ2 =0.387,P>0.05),but the CD31 gene Leu125Val polymorphism of the 2 groups had significant difference (χ2=10.875,P〈0.05).The relative risk of HCC of Val allele was 1.457 times that of the Leu allele carriers(OR=1.457,95%CI:1.187~2.186,P=0.001);Serum level of CD31 of Val allele carriers was significantly higher than that of no carriers (χ2 =10.534,P〈0.05).Genotype analysis showed that CD31 gene Leu125Val,Asn563Ser and Gly670Arg polymorphisms had strong linkage disequilibrium ,the Val-Ser-Arg haplotype was associated with a high risk of HCC as compared with the healthy patients (OR=1.487,95%CI:1.092-2.038,P=0.011).Conclusion CD31 gene Leu125Val polymorphism and its Val-Ser-Arg haplo-type are associated with HCC ,and Val allele is very likely an important genetic susceptibility gene for HCC .CD31 Val allele carri-ers may subject to higher risk of HCC with enhanced CD 31 expression .
