川芎嗪对乏氧人肺癌细胞β1-整合素及钙依赖黏附素表达的影响 被引量:2

摘要 目的:探讨川芎嗪对乏氧人巨细胞肺癌(PG)细胞表达β1-整合素和钙依赖黏附素的影响。方法:PG细胞复苏,分为川芎嗪高、中、低剂量组及顺铂组、阴性对照组,置常氧和低氧环境培养,检测E-cadherin(CD324)、β1-整合素(CD29)的表达。结果:PG细胞CD324常氧组表达大于低氧组;在常氧和低氧环境中,各实验组CD324表达均大于对照组;常氧组CD29表达低于低氧组。结论:在乏氧环境下PG细胞有更高的转移能力,川芎嗪能够抑制PG细胞的转移能力。 Objective: To investigate the effect of Tetramethylpyrazine( TMP) on the expression of β1-integrin( CD29)and E-cadherin( CD324) on the surface of cell PG cultured during normoxia and hypoxia. Methods: The revival cell PG was divided into three dosages( high,medium and low) group of Tetramethylpyrazine,DDP group and negative control group.All of five groups were cultured separately in the normal and hypoxic environment. The expression of CD324 and CD29 on the surface of PG cell were detected by the flow cytometry 48 h later. Results: The positive rate of PG cells ’CD324expression of normal oxygen group is higher than hypoxic group( P 〈 0. 01). In normoxic and hypoxic microenvironment,the expression rate of all experimental group is higher than control group( P 〈 0. 05). The intensity of PG cells’ CD324 expression of normal oxygen group is lower than hypoxic group( P 〈 0. 01). Conclusion: PG cells have a higher metastatic ability in the hypoxic microenvironment. In the normoxic and hypoxic microenvironment,tetramethylpyrazine can inhibit metastasis ability of PG cells through regulating the metastasis-associated adhesion molecules.
