摘要
将类转录激活因子效应物核酸酶(Transcription activator-like effector nuclease,TALEN)打靶技术运用到水稻叶绿体转化体系中,有可能提高外源基因在水稻叶绿体基因组中的整合效率。根据叶绿体表达载体上的同源重组序列,构建了水稻叶绿体TALEN载体,经分子检测和DNA测序分析证明载体构建成功;将该载体转化大肠杆菌,载体上携带的EGFP、hpt和Fok基因在大肠杆菌中得到成功表达,说明构建的水稻叶绿体TALEN载体能够在原核系统中有效表达。
The TALEN targeting technology was applied to rice chloroplast transformation system to improve the integration efficiency of foreign genes in rice chloroplast genome. A TALEN vector for rice chloroplast was constructed according to the homologous recombination sequence of chloroplast expression vector. Molecular analysis and DNA sequencing showed that the vector was successfully constructed. The genes of EGFP,hpt and Fok were expressed in E. coli transformed with the TALEN vector,indicating that the TALEN vector for rice chloroplast was effectively expressed in E. coli.
