摘要
以交联法、交联包埋法和包埋交联法固定化Alealase2.4L碱性酶,对3种方法制备的固定化酶的酶活回收率和特性进行了对比。结果表明,交联法固定化酶的酶活回收率最高,达到45.7%。交联法、交联包埋法和包埋交联法制备的固定化酶的最适温度分别升高5、10和10℃,最适pH分别降低1、2和2个单位,Km值分别升高了1.65、2.18和2.34倍,热稳定性和储藏稳定性得到很大提高,交联包埋法固定化酶的稳定性最高。
Alcalase2.4L alkaline protease was immobilized by the means of chemical cross -linking, crosslinking -entrapment and entrapment - crosslinking. The enzyme recovery of protease and partial properties of Alcalase2.4L alkaline protease immobilized by different methods and free enzyme were investigated. The results showed: the enzyme recovery of protease immobilized by chemical cross- linking was highest, reached 45.7%. The optimal temperature of the immobilized proteases by cross- linking, crosslinking- entrapment and entrapment - crosslinking was up 5, 10 and 10℃, while the optimal pH declined 1, 1 and 2, respectively. The kinetic parameter, Kin, for immobilized enzymes were 1.65, 2.18 and 2.34 times free enzyme. The thermal, and storage stabilities of the enzyme were improved greatly after they were immobilized, especially the protease immobilized by the mean of crosslinking - entrapment.
