鼻咽癌中NF-κB对Hedgehog通路基因表达的影响 被引量:1

摘要 目的 Hedgehog通路与NF-κB均被发现与多种癌症相关,本研究初步探讨在鼻咽癌中两者之间的相关性,从而为靶点治疗等临床应用提供可靠的实验依据。方法提取12例鼻咽癌、鼻咽炎患者病例样本总RNA反转录得m RNA的c DNA进行荧光定量PCR,通过统计相关系数得出Hedgehog通路与NF-κB相关性;利用不同浓度NF-κB抑制剂PDTC(50~400μmol/L)作用24 h鼻咽癌细胞株CNE1,以CCK8法检测PDTC对CNE1增殖的影响;并对受不同浓度PDTC影响下CNE1的Hedgehog通路与NF-κB基因表达进行荧光定量PCR测定。结果鼻咽癌、鼻咽炎病例样本中Hedgehog通路基因Gli、PTCH、SMO与NF-κB相关系数大于0.8,呈现较强的相关性;PDTC对CNE1有显著的增殖抑制,而且显著抑制NF-κB及Hedgehog通路各基因的表达。结论鼻咽癌中NF-κB与Hedgehog有较强的相关性,NF-κB抑制剂PDTC能显著抑制鼻咽癌细胞株CNE1增殖以及NF-κB、Hedgehog信号通路基因表达。 Objective Previous researches suggested that Hedgehog signaling pathway and NF-κB were involved in various cancers.This study investigated the correlation between Hedgehog signaling pathway and NF-κB,which maybe provide a reliable evidence for clinical treatment.Methods Total RNA was isolated from patients with nasopharyngeal cancer and nasopharyngitis and converted to c DNA.Real-time PCR was performed to detect the expression level of relevant genes.Then the correlation between Hedgehog signaling pathway and NF-κB was evaluated by analyzing relevant data;Nasopharyngeal cancer cell line CNE1 was treated with PDTC(NF-κB inhibitor) for 24 h in different concentration,ranging from 50 μmol / L to 400 μmol / L.The effect of PDTC on the proliferation of CNE1 was investigated using CCK8 and the expression of genes involved in Hedgehog signaling pathway as well as NF-κB was detected by Real-time PCR.Results The correlation coefficient between NF-κB and genes involved in Hedgehog signaling pathway such as Gli,PTCH and SMO was higher than 0.8,which indicated that there was a strong correlation between them;PDTC had significant inhibition to the proliferation of CNE1,and had significant inhibition to the expression of NF-κB and Hedgehog genes.Conclusion NF-κB was highly related to Hedgehog signaling pathway.Furthermore,PDTC,a NF-κB inhibitor,can greatly reduce the proliferation of CNE1 and the expression of NF-κB and genes involved in Hedgehog signaling pathway.
