摘要
目的:研究三黄汤的血清药物化学。方法:建立三黄汤及各单味药(茵陈、黄芩、黄柏、甘草、炙大黄)、缺味药水提物与大鼠ig给药后血清成分的高效液相(HPLC)指纹色谱图;分析三黄汤及各单味药、缺味药水提物给药不同时间后的入血成分、相对含量、来源及其代谢产物。结果:建立的三黄汤体外指纹图谱标记出44个主要成分;大鼠ig给予三黄汤后血清指纹图谱标记出32个主要入血成分,分别为源于茵陈的包括绿原酸在内的4个原型成分及1个绿原酸代谢产物,源于黄芩的包括黄芩苷在内的16个原型成分及2个代谢产物,源于黄柏的盐酸小檗碱,源于甘草的4个原型成分,源于大黄的大黄酸,以及未知来源的3个代谢产物。随着给药时间的延长,绿原酸和黄芩苷等原型成分的相对含量逐渐减少,而其代谢产物逐渐增加。结论:血中移行成分主要来源于茵陈和黄芩。
OBJECTIVE:To investigate the serum pharmacochemistry of Shanhuang decoction(SHD). METHODS:HPLC fingerprints of serum components of rats which were ig given the water extracts from SHD,single herb medicines(Artemisiae Scopariae Herba,Scutellaria baicalensis,Phellodendron chinense,Glycyrrhizae Radix et Rhizoma,Rhei Radix et Rhizoma)and the medicines without some herb were established. The components in blood,relative contents,sources and metabolites which were detected at different time after the administration of the above-mentioned water extracts were analyzed. RESULTS:44 main components were marked in the fingerprint of SHD in vitro. After the ig administration of SHD,32 main components into blood were marked in the serum fingerprint,such as 4 prototype components including chlorogenic acid and 1 metabolite from Artemisiae Scopariae Herba,16 prototype components including baicalin and 2 metabolites from S. baicalensis,berberine hydrochloride from P. chinense,4 prototype components from Glycyrrhizae Radix et Rhizoma,rhein from Rhei Radix et Rhizoma,and 3 metabolites of unknown origin. The contents of the prototype components such as chlorogenic acid and baicalin were gradually decreased by the increasing of administration time,and more and more metabolites would be produced. CONCLUSIONS:The transitional components in blood are mainly derived from A. Thunb and S. baicalensis.
