摘要 目的:研究胃癌组织中性别决定相关基因簇2(SOX2)与转化生长因子β1(TGF-β1)、Smad3的表达情况及与胃癌组织临床病理因素之间的关系,并探讨三者之间的相关性。方法:采用免疫组织化学法标记SOX2、TGF-β1及Smad3,检测其在正常胃组织和胃癌组织中的蛋白表达情况,并结合临床病理学资料对实验数据进行统计学分析。结果:SOX2、Smad3蛋白在胃癌组的积分光密度(IOD)值低于正常胃组织(P <0.05),TGF-β1在胃癌组的IOD值高于正常胃组织(P <0.05)。 TGF-β1 IOD值的高表达多见于低分化、有淋巴结转移及Ⅲ~Ⅳ期的胃癌组中(P <0.05);SOX2、Smad3 IOD值的高表达多见于高-中分化、无淋巴结转移及Ⅰ+Ⅱ期胃癌组中(P <0.05);SOX2、TGF-β1和Smad3蛋白在胃癌组织中的IOD值与患病年龄、性别无关(P >0.05)。在胃癌中TGF-β1与Smad3之间呈负相关性;SOX2与Smad3之间呈正相关性;TGF-β1与SOX2之间呈负相关性。结论:SOX2、TGF-β1及Smad3蛋白的异常表达与胃癌的发生、发展和临床病理特征有关。 Objective To investigate the expression of SOX2,TGF-β1 and Smad3 in gastric cancer and their relationships with clinical pathological characteristics of gastric cancer. Methods Immunohistochemical staining was used to detect the expression of SOX2, TGF-β1 and Smad3 expression and the clinical pathological characteristics of gastric cancer were analyzed. Results Integrated optical density (IOD) values from the positive expression of SOX2, Smad3 in gastric cancer group were all significantly lower than normal gastric tissues (P 0.05). There was positive correlation between SOX2 and Smad3 expression in gastric cancer tissues, and negative correlation between TGF-β1 and Smad3 in gastric cancer. Conclusions Abnormal expression of SOX2, TGF-β1 and Smad3 protein is related to the onset, development and clinicopathologic features of gastric cancer.
