摘要
目的建立测定卡托普利缓释片中卡托普利缓释片含量的高效液相色谱法。方法色谱柱为Agilent Eclipse XDB-C18柱（250 mm×4.6 mm,5μm）,0.01 mol/L磷酸二氢铵水溶液（用磷酸调p H至3.0±0.05）-甲醇-乙腈（65∶25∶15）为流动相,流速为1.0 m L/min,检测波长为215 nm。结果卡托普利质量浓度在0.2~2.0μg/m L范围内与峰面积线性关系良好（r=0.999 9）,平均回收率为99.34%,RSD=0.52%（n=9）。结论该方法样品分离度效果好,测定结果准确、可靠、重复性好,可用于卡托普利缓释片中卡托普利的含量测定。
Objective To establish a high performance liquid chromatography（ HPLC） method for the direct determination of the captopril content in Captopril Sustained- Release Tatlets. Methods The chromatographic column was the Agilent Eclipse XDB- C18 column（ 250 mm × 4. 6 mm,5 μm） with 0. 01 mol / L ammonium dihydrogen phosphate aqueous solution（ adjusting p H to 3. 0 ± 0. 05 with phosphoric acid）- methanol- acetonitrile（ 65 ∶ 25 ∶ 15） as the mobile phase,the flow rate was 1. 0 m L / min and the detection wavelength was 215 nm. Results The mass concentration of captopril in the range of 0. 2- 2. 0 μg / m L showed the good linear relation with the peak area（ r = 0. 999 9）,the averaga recovery rate was 99. 34%,RSD = 0. 52%（ n = 9）. Conclusion The separation obtained by adopting the established method is good and the detection results are accurate and reliable with good reproducibility.
