山茱萸环烯醚萜苷促进成体大鼠海马神经干细胞增殖和分化的影响 被引量:11

Effects of cornel iridoid glycoside on neurogenesis in adult rats

摘要 目的:观察山茱萸环烯醚萜苷(CIG)对成体大鼠神经发生的影响。方法:成年Wistar大鼠,分为正常对照组、CIG 60 mg·kg-1组、CIG 180 mg·kg-1组,灌胃给药,正常对照组灌蒸馏水,给药d 26开始,腹腔注射Brd U到大鼠体内连续3 d,bid,分别于24 h和28 d后处死动物取脑。应用免疫组织荧光法观察大鼠海马齿状回Brdu标记的神经干细胞,用免疫组织荧光双标染色法检测Brdu/Map-2标记的新生神经元。结果:Brd U标记4 d后,与正常对照组相比,CIG 180 mg·kg-1组大鼠海马齿状回的Brd U阳性细胞数显著增加(P<0.01);Brd U标记28 d后,CIG 60 mg·kg-1组和CIG 180 mg·kg-1组Brd U/MAP-2阳性细胞数明显增多(P<0.05,P<0.01)。结论:CIG对成年大鼠海马齿状回内源性神经干细胞的增殖及分化具有促进作用。 Objective: To investigate the effects of cornel iridoid glycoside( CIG) on neurogenesis in adult rats. Methods: Adult Wistar rats were randomly divided into groups,and treated with i. g. CIG( 60 and 180 mg·kg- 1) or distilled water( normal control) for 26 days. Thereafter,Brd U was intraperitoneally injected into the rat,2 times a day for 3 days. The brain was isolated at 24 h and 28 d,respectively. Immunofluorescence assay was used to detect the expression of Brd U-labeled neural stem cells( NSCs) in the hippocampal dentate gyrus. Double immunofluorescence labeling was applied to detect Brd U / Map-2 labeled mature neurons differentiated from NSCs. Results: Compared with the normal control,the number of Brd U-labeled cells in hippocampal dentate gyrus was significantly increased in CIG-treated rats at 4 days after Brd U labeling( P〈0. 01). The number of Brd U / MAP-2positive cells was obviously elevated in CIG-treated rats at 28 days after Brd U labeling( P〈0. 05 and P〈0. 01).Conclusion: CIG is able to promote the proliferations and differentiation of endogenous NSCs in hippocampal dentate gyrus of adult rats.
