摘要
目的:研究麻黄附子细辛汤与头孢唑林联合应用于耐甲氧西林金葡菌时对头孢唑林的增效作用,为研究中药对抗生素增效的作用机制和筛选活性成分奠定实验基础。方法:将麻黄附子细辛汤复方、中药复方加头孢唑林、头孢唑林分别加入培养基中,同时制作未加药物的空白培养基作为空白组和阴性组,以及单纯加入替加环素的培养基作为阳性组,分别接种耐甲氧西林金葡菌,空白组不接种细菌,每12 h取出计数,计算活菌数;同时使用连续稀释法检测各自最小抑菌浓度(MIC)及最低杀菌浓度(MBC);并且用纸片法检测抑菌圈。结果:(1)中药复方组12 h时发现细菌逐渐生长,直至培养120 h后细菌浓度才逐渐下降,直至培养180 h时细菌浓度保持不变,头孢唑林组培养12 h时细菌数逐渐减少,到60 h时细菌减少的趋势停止,中药复方联合头孢唑林组培养12 h时细菌数逐渐减少,并在培养144 h时各组细菌菌无生长迹象,替加环素药物干预后细菌数急剧下降,但是直到观察结束仍有部分细菌存活,阴性组细菌随着培养时间的逐渐延长呈现缓慢生长趋势,空白组无细菌生长。(2)麻黄附子细辛汤对耐甲氧西林金葡菌(MIC=2.5 mg/m L,MBC=5 mg/m L);头孢唑林对耐甲氧西林金葡菌(MIC=1.25 mg/m L,MBC=5 mg/m L);中药复方联合头孢唑林对耐甲氧西林金葡菌(MIC=0.312 mg/m L,MBC=1.25mg/m L);替加环素对耐甲氧西林金葡菌(MIC=0.156 mg/m L,MBC=0.625 mg/m L)。(3)中药复方联合头孢唑林抑菌环直径和阳性对照组相近,其次是头孢唑林组,中药单独使用的抑菌环直径小于头孢唑林组.麻黄附子细辛汤与头孢唑林联合用药对耐甲氧西林金葡菌的抗菌效应增强,麻黄附子细辛汤与头孢唑林联合用药的抗茵作用表现为协同作用(FIC=0.59)。结论:麻黄附子细辛汤联合抗生素具有更明显的抗菌效应,可能与它激活耐药细菌生长有关。
Objective: To study the synergistic effect of ephedra monkshood asarum decoction combined antibiotic,for studying the mechanism of traditional Chinese medicine synergistic on antibiotics and laying the experimental foundation for screening of active ingredients. Methods: The ephedra monkshood asarum decoction, the compound Chinese medicine combined cefazolin, cefazolin were added in the culture medium respectively,simultaneously made the blank culture medium as blank group and negative group without drugs,and culture medium only added Tigecycline as positive group,overnight cultured at 37 ℃ after inoculation of methicillin-resistant Staphylocus aureus respectively,the blank group didn't inoculate any bacterium,then took out every 12 hours for counting,calculate the number of viable counts,at the same time,detected their minimal inhibitory concentration( MIC) and minimum bactericidal concentration( MBC) by continuous dilution method,and detected inhibition zone by paper slip method. Results:( 1)It was found that bacteria grew gradually in the traditional Chinese medicine compound group at 12 h,after cultured for 120 h,the bacteria concentration gradually decreased,until cultured for 180 h,the bacteria concentration remained the same,bacteria count reduced gradually in cefazolin group when cultured 12 h,the trend of bacteria reduction ended at 60 h,when cultured 12 h the bacteria count of traditional Chinese medicine compound combined cefazolin group reduced gradually,when cultured for 144 h,all groups of bacteria showed no sign of growth,bacterial count fell sharply after Tigecycline intervation,but some bacteria survive until the end of the observation,the bacteria of negative group showed a trend of slow growth along with culture time gradually extend,the blank group didn' t have bacterium growth.( 2) The ephedra monkshood asarum decoction vs. methicillin-resistant Staphylocus aureus( MIC =2. 5 mg / m L,MBC = 5 mg / m L); Cefazolin vs. methicillin-resistant Staphylocus aureus( M
