旋覆花中倍半萜内酯类成分提取工艺的优化及体外抗肿瘤药效研究 被引量:5

摘要 目的采用正交试验设计考察旋覆花中倍半萜内酯类提取的影响因素,优选其最佳提取工艺,并结合体外抗肿瘤药效学实验,考察其抗肿瘤的药理活性,为旋覆花的开发和利用提供理论依据。方法采用正交试验设计以1-O-乙酰基大花旋覆花内酯为考察指标,对影响倍半萜内酯类成分的提取工艺的4个因素乙醇浓度、提取时间、提取次数、乙醇用量进行考察,优选最佳提取工艺。采用四唑盐MTT比色法检测对人肝癌细胞生长的影响,探讨其体外抗肿瘤细胞增殖作用的规律。结果最佳提取工艺条件为15倍量80%乙醇提取3次,每次2 h,其抗肿瘤增殖作用呈现出良好的时间-剂量-效应关系。结论本实验优选的最佳提取工艺及其体外抗肿瘤作用为旋覆花的研究提供实验依据和理论基础。 objective To inspect the influencing factors of extracting sesquiterpene lactones from Flos Inulae,and op-timize the extraction process by orthogonal test. To investigate the pharmacological activity of anti-cancer effect combining with the experiments of antineoplastic pharmacodynamics in vitro,and provide references and theoretical basis for the devel-opment and utilization of Flos Inulae. Methods Four main factors of ethanol concentration,extraction time,times and etha-nol amount were investigated to screen the optimum extraction process with ABL as investigating index by orthogonal experi-mental design. This experiment used MTT assay to evaluate the antitumor activities of sesquiterpene lactones to the cell line of HepG2 cells,and to explore the rule of the antitumor activities. Results The optimum conditions for the extraction of Flos Inulae were:15 times amount of 80% ethanol,3 times reflux extraction,2 hour each time. The sesquiterpene lactones suppressed the proliferation of HepG2 cells,it had a good does-time-effect relations. Conclusion The experiment was designed to optimize the extraction process of Flos Inulae and the antitumor activities of sesquiterpene lactones in vitro,it provided the basis of experiment and theoretical basis for Flos Inulae.
