摘要
【目的】建立鸡蛋花环烯醚萜含量测定方法,优选其提取工艺。【方法】采用紫外分光光度法,以梓醇为对照品,经2,4-二硝基苯肼显色后,在463 nm处测定其含量,并利用单因素结合正交设计优选最佳的提取工艺。【结果】该显色方法稳定,梓醇浓度在0.0220~0.0882 mg/m L范围内线性关系良好,回归方程为Y=10.18 X-0.020,R2=0.999,平均回收率为99.72%,相对标准偏差sR=2.9%(N=6)。优化的工艺条件是乙醇浓度60%(体积分数),液料比25∶1,超声提取60 min。【结论】该方法准确,简单,稳定性良好,工艺稳定可行,可为鸡蛋花生产工艺及质量控制提供参考。
Objective To set up a method for the determination of the total iridoid in Flos Plumeriae Acutifoliae,and to optimize the extraction process. Methods With catalpol as the reference, the total iridoid in Flos Plumeriae Acutifoliae was determined by ultraviolet( UV) spectrophotometry at wavelength of 463 nm after coloration with 2, 4-dinitro-phenylhydrazine. The experiment was carried out by orthogonal design, and single factor analysis was used to optimize the extraction process. Results The results of the coloration were stable.Catalpol showed good linearity in the range of 0.022 0 ~ 0.088 2 mg/m L( R2=0.999), regression equation was Y=10.18 X-0.020, the average recovery was 99.72%, and relative standard deviation(RSD) was 2.9%(N=6).The optimized proces condition was ethanol concentration being 60%, ultrasonic extraction for 60 minutes and the liquid-solid ratio being 25 ∶1. Conclusion The established method is accurate, simple and stable. The extraction process is stable and feasible, and can supply evidence for the quality control and production process of Flos Plumeriae Acutifoliae.
