狗肝菜多糖对二甲基亚硝胺诱导的肝纤维化大鼠的影响 被引量:12

摘要 目的:以二甲基亚硝胺诱导的大鼠肝纤维化为模型,研究狗肝菜多糖对其拮抗作用及其机制。方法:将96只SD大鼠随机分为空白对照组、模型组、狗肝菜多糖(300 mg/kg、200 mg/kg、100 mg/kg)组和秋水仙碱组(0.12 mg/kg)。除正常对照组以外,其余各组大鼠均腹腔注射0.5%二甲基亚硝胺溶液(1.5 ml/kg),隔天1次,连续5周。5周末摘大鼠眼球取血和收取肝脏,检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、丙二醛(MDA)和超氧化物歧化酶(SOD);检测肝组织中透明质酸(HA)、层黏连蛋白(LN)、Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(Ⅳ-C)、肿瘤坏死因子α(TNF-α)、白介素6(IL-6)和转化生长因子β1(TGF-β1)水平;并通过苏木精-伊红染色(HE染色)进行肝组织病理学检查。结果:狗肝菜多糖(300 mg/kg、200 mg/kg、100 mg/kg)组和秋水仙碱(0.12 mg/kg)组大鼠血清或组织中ALT、AST、MDA、SOD、HA,LN,PCⅢ,Ⅳ-C、TNF-α、IL-6和TGF-β1水平或含量均显著低于模型组;病理切片显示狗肝菜多糖组和秋水仙碱组大鼠肝组织炎症坏死程度和肝纤维化病变程度较模型组明显减轻。结论:狗肝菜多糖具有抗DMN诱导的大鼠肝纤维化的作用,其机制可能与保肝、抗氧化、抗炎和降低TGF-β1表达有关。 Objective: Study on the antagonistic effects and and mechanism of Diclipterachinensis polysaccharide on liver fibrosis in rats induced by Dimethylnitrosamin. Methods: 96 SD rats were randomly divided into blank control group, model group, Dicliptem chinensis polysaccharide (300 mg/kg, 200 mg/kg, 100 mg/kg) groups and colchicines(0.12 mg/kg) group. Except the normal control group, the other rats injected 0.5% Dimethylnitmsamin solution (1.5ml/kg) through abdominal cavity, every two days for 1 times in 5 weeks. Blood was taken by picking off eyeball and liver was collected at 5 weekend in the experimental, The ALT, AST, MDA ,SOD in serum and HA, LN, PCIH, IV-C, TNF-a, IL-6 , TGF-β1 were detected in liver tissue of rats. Liver histopathological examination by hematoxylin eosin staining ( HE staining). Results: The Dieliptera chinensis polysaccharide (300 mg/kg,200 mg/kg, 100 mg/kg) groups and colchicines (0.12 mg/kg) group of the ALT, AST, MDA, HA, LN, PC III, IV -C, TNF-a,IL-6 and TGF-β1 levels were significantly lower and SOD was higher than the model groups ( P 〈 0.05 or P 〈 0.01 ) ;The Flammation and necrosis and the degree of liver fibrosis was significantly reduced of the drug therapy compared with model group. Conclusion: Dicliptera chinensis polysaccharide has anti DMN induced liver fibrosis rats, its mechanism may be related to the pmtectliver, antioxidant,anti-inflammatory and decrease the expression of TGF-β1.
