山茱萸总苷对急性缺氧乳鼠心肌细胞凋亡通路的影响 被引量:6

Effect of total glycosides from Cornus officinalis on myocardial apoptosis pathway in suckling mouse with acute hypoxia

摘要 目的:探讨山茱萸总苷对急性缺氧乳鼠心肌细胞凋亡通路的影响。方法:原代培养乳鼠的心肌细胞,采用液体石蜡封闭法建立缺血缺氧模型,并将原代培养3天的心肌细胞分为正常组、缺氧组(1h、2h、3h、5h)及药物治疗组,药物浓度采用山茱萸总苷三个剂量组,免疫组化法检测各组cleaved caspase-9表达。结果:与对照组相比,缺氧各组caspase-9表达增加(P<0.01),并且随着时间延长,阳性表达的比例增加,5h达到最高,给予山茱萸总苷干预之后,治疗组每个时间点cleaved caspase-9表达都有所下降,随着山茱萸总苷剂量的增加,cleaved caspase-9表达逐渐下降,与缺氧组相比,没有统计学意义,但0.049g/L和0.49g/L组差异有统计学意义(P<0.01)。山茱萸总苷起效剂量为0.049g/L,剂量范围为0.049g/L^0.49g/L。结论:山茱萸总苷可以抑制cleaved caspase-9的表达,可能是通过阻断心肌细胞凋亡线粒体通路去实现对心肌细胞起保护作用。 Objective:To investigate the effects of total glycosides from Comus officinalis on myocardial apoptosis pathway in suckling mouse with a- cute hypoxia. Methods: Myocardiocytes of neonatal rats were incubated with liquid paraffin closed method to establish the ischemic anoxia model. The generation of 3 days training myocardial cells were divided to normal group,hypoxia group (1h,2h,3h,Sh) and drug group. The concentration was divided to 0.1 g/L, 1 g/L, and 10g/L three group. Immunohistochemical method was used to test the easpase-9. suits: Compared with blank, the expression of cleaved caspase-9 of every oxygen-poor group is increased, and the difference was statistical significance (P 〈0.01). As the extension of time, the positive expression of cleaved caspase-9 was increasing, and the peak was in 5h. With the intervention of total glycosides from Comus ofiqcinalis ,each time of cleaved caspase-9 in drug group decreased. As the increasing of total glycosides from Comus ottleinalis, the expression of cleaved caspase-9 was decreasing in each drug group. And compared with oxygen- poor group, the 0. 0049g/L group was no statistical significance ( P〉0.05 ), but the 0.049g/L and 0.49g/L group had the statistical signifi- cance ( P 〈0.01 ). The effective dose of total glycosides from Comus ottlcinalis was 0.049g/L, and the range of total glycosides from Comus oitlcinalis was between 0.049g/L to 0.49g/L. Conclusion: Total glycosides from Comus officinalis could inhibit the expression of cleaved caspase-9. The mechanism of that may be by blocking the myocardial mitochondria apoptosis pathway to reach the protection of myocardial cells.
