摘要
目的观察参芪益心方对阿霉素所致慢性心力衰竭(CHF)大鼠心肌细胞凋亡的影响,探讨益气温阳,活血利水法对心肌细胞凋亡的干预效应及机制。方法将90只Wistar雄性大鼠随机平均分为正常对照组、模型组、依那普利对照组(西药对照组)、芪苈强心对照组(中药对照组)、参芪益心方低剂量组、参芪益心方高剂量组。阿霉素复制CHF大鼠模型,造模大鼠药物干预4周。TUNEL法测定心肌细胞凋亡率;RT-PCR法检测心肌组织中Caspase-3及Caspase-9 mRNA的表达;Western blot法检测心肌组织中Caspase-3和Caspase-9蛋白的表达。结果各治疗组药物干预后均可降低CHF大鼠心肌组织Caspase-3、Caspase-9 mRNA及蛋白表达,除参芪益心方低剂量组外,各治疗组与模型组比较,差异均有统计学意义(P<0.05);参芪益心方高剂量组与中、西药对照组比较,差异无统计学意义(P>0.05)。结论参芪益心方可以降低阿霉素所致CHF大鼠死亡率,下调心肌组织Caspase-3及Caspase-9表达,其抑制心肌细胞凋亡的作用可能是改善心功能的作用机制。
Objective To observe the impacts of Shenqi Yixin Formula on myocardial apoptosis in the rats of chronic heart failure(CHF) induced by Doxorubicin and explore the intervention effect and mecha- nism on myocardial apoptosis treated with the therapy for benefiting qi, warming yang, activating blood circu- lation and promoting water metabolism. Methods Ninety Wistar male rats were randomized averagely into a normal control group, a model group, an Enalapril control group, a Qili Qiangxin control group, a Shenqi Yixin Formula low -dose group( formula low -dose group)and a Shenqi Yixin Formula high -dose group( formula high - dose group). Doxorubicin was used to duplicate CHF rat model. The medicinal intervention was given in the model rats for 4 weeks. TUNEL method was applied to determine myocardial apoptosis, RT - PCR assay was to determine mRNA expressions of Caspase - 3 and Caspase - 9 in myocardial tissue and Western blot method was to determine the protein expressions of Caspase - 3 and Caspase - 9. Results After the medici- nal intervention, the mRNA and protein expressions of Caspase - 3 and Caspase - 9 were all reduced in myo- cardial tissue in CHF rats of each treatment group. Except in the formula low - dose group, as compared with the model group, the differences were significant in the rest treatment groups (P 〈 0.05 ). Compared with the Chinese medicine control groups and western medicine group, the differences were not significant in the for- mula high -dose group( P 〉 0.05 ). Conclusion Shenqi Yixin Formula can reduce the death rate of CHF rats induced by Doxorubicin and down - regulate the expressions of Caspase - 3 and Caspase - 9 in myocardial tissue. The inhibition of the formula on myocardial apoptosis is possibly the mechanism on the improvement of heart function.
