摘要
通过透射电子显微技术对羊口疮病毒(ORFV)YX强毒株感染羊胚胎鼻甲细胞(OFTu)的细胞超微结构和病毒形态发生学进行研究.结果显示,成熟的病毒粒子呈椭圆形,有囊膜,大小约为150 nm×250 nm,可见两面凹陷呈哑铃形的核酸芯髓和2个侧小体结构.病毒在胞浆中复制,在高尔基体形成囊膜,最后病毒粒子通过细胞膜出芽和细胞裂解方式释放病毒粒子.感染细胞超微结构主要表现为:内质网扩张呈囊、池内分离;线粒体增生、嵴肿胀、变暗、模糊不清,最后空泡化;高尔基体出芽、扩张;细胞核溶解呈早期凋亡现象,胞浆出芽,最后整个细胞裂解、破碎.
The morphogenesis of orf virus（ ORFV） virulent strain YX in OFTu cells and the ultrastructural changes of host cells were observed and analyzed by using transmission electron microscopic technique. The mature virion particles were enveloped,ovoid in shape with diameter of 250 × 150 nm. Dumbbell shape DNA core and two corpuscles were observed in some virus particles. Viruses replicated and bred in the cytoplasm and gained capsule membrane from the Golgi. The mature viruses were released from infected cells by budding or cell broken. The main ultrastructural changes of the infected cells were as follows： expanding endoplasmic reticulum like a capsule,with pool separation; mitochondria showing hyperplasia,swell,dark,vague,finally demonstrating vacuolation; Golgi budding,enlarging. Cell core dissolving showing apoptosis changes,cytoplasm budding,in the end infected cells were destroyed by the virus,dissolving.
