猪流行性腹泻病毒感染Vero细胞不同时间的超薄切片电镜观察 被引量:2

Observation of porcine epidemic diarrhea virus infecting Vero cells by electron microscopy with ultrathin sections at different time

摘要 为了研究猪流行性腹泻病毒(PEDV)体外培养的生物学特性,试验采集南京市某猪场疑似病毒性腹泻的病死仔猪病料,运用RT-PCR法进行PEDV初步鉴定,将鉴定结果为阳性的病料接种Vero细胞进行培养,并通过透射电镜对PEDV感染Vero细胞不同时间进行细胞超薄切片电镜观察。结果表明:病毒呈球形或椭圆形,其直径为60~80 nm,部分呈中空状,多呈致密核芯。病毒粒子成堆或成串地排列在细胞浆内,随感染时间的延长病毒粒子逐渐增多,部分病毒粒子聚集在粗面内质网周围,细胞浆空泡增多,线粒体增生、肿胀、空泡化。在感染最后阶段,仍见大量病毒粒子聚集在胞浆内,细胞裂解破碎。 To study the biological characteristics of Porcine epidemic diarrhea virus (PEDV) cultured in vitro, the pathological materials of dead, diseased piglets with suspected viral diarrhea were collected from a pig farm in Nanjing. The RT - PCR assay was used for preliminary i- dentification of PEDV, the pathological materials identified as positive, were used to inoculate Vere cells for the culture. The observation using electron microscopy with ultrathin sections for PEDV infecting Vero cells was carried out through transmission electron microscope. The results showed that the viral particles were spherical or oval - shaped, the viral diameters were in the range of 60 to 80 nm. Some of viral particles were hollow, most were dense - core in morphology. The viral particles arranged in piles or clusters within the cytoplasm and increased gradually with prolonged infection. Some of viral particles gathered around the rough endoplasmic reticulum. The number of cytoplasmic vacuoles increased, and the hyperplasia, swelling and vacuolization occurred in the mitochondria. In the final stage of infection, a large number of viral particles accumu- lated in the cytoplasm, and the whole cell lysis and broken were observed.
