摘要
目的评估氧诱导视网膜病变(oxygen induced retinopathy,OIR)中成纤维母细胞生长因子2(fibroblast growth factor-2 FGF2)的时空变化并探讨其作用机制。方法实验研究。对2012年1月至2013年8月在成都中医药大学附属医院眼科,随机化将C57BL/6J幼鼠分实验组和对照组,实验组制作OIR模型,P17取各组视网膜行HE染色及异硫氰酸荧光素(fluorescein isothiacyanate—dextran,FITC—dextran)眶后注射视网膜铺片确定OIR模型建立;P13、P15、P17取视网膜,WEST BLOT检测FGF:时空表达变化。结果OIR模型建立成功,P13、P15、P17实验组FGF2表达呈现上升趋势,明显高于对照组。结论OIR可能通过增加FGF2表达抑制氧诱导视网膜病变。
Objective To evaluate the spatial and temporal variations of fibroblast growth factor 2 (FGF2) in oxygen induced retinopathy, and to explore its action mechanism. Methods C57BL / 6J pups were used and assigned to experimental group and control group, experimental group was OIR model. For P17, use the pave piece of retina with HE staining and retrobulbar injection of fluo- rescein isothiocyanate-dextran injection to create the OIR model; P13, P15, P17 to check the retinal FGF2 expression of time and space change by WEST BLOT. Results OIR model was constructed successful, FGF2 expression showed a rising trend in the P13, P15, P17 experimental group, signifi- cantly higher than the control group. Conclusions OIR may inhibit of the oxygen induced retinopa- thy by increasing FGF2 expression.
