摘要
【目的】研究京尼平苷酸(geniposide-acid,GA)对巨噬细胞培养上清液刺激滑膜细胞RSC-364增殖及其分泌致炎因子的影响。【方法】佛波酯(phorbolmyristate acetate,PMA)诱导THP-1细胞分化为巨噬细胞,加入脂多糖(lipopolysaccharide,LPS)刺激后获取培养上清液,将RSC-364细胞分为对照组,模型组,甲氨蝶呤组(1×10-6mol/L),GA高(1×10-5mol/L)、中(1×10-6mol/L)和低(1×10-7mol/L)剂量组,进行MTT细胞增殖试验,流式细胞术检测细胞周期,ELISA法测定细胞上清液肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α),白细胞介素-1β(interleukin-1β,IL-1β)水平。【结果】GA能剂量依赖性抑制RSC-364细胞增殖及IL-1β、TNF-α分泌,其中高剂量组作用最显著,与模型组相比,1×10-5mol/L GA能明显抑制细胞增殖(24、48和72 h抑制率分别为25%,21%和20%,P<0.01),明显增加细胞周期G1期细胞数[(79.20±0.55)%vs(69.22±0.33)%,P<0.01],明显降低细胞外液IL-1β[(14.35±0.12)vs(40.55±0.61)ng/L,P<0.01]和TNF-α[(11.92±0.71)vs(40.45±0.38)ng/L,P<0.01]含量。【结论】GA能显著抑制巨噬细胞培养上清液刺激下的RSC-364细胞增殖及炎症细胞因子TNF-α和IL-1β分泌。
【Objective】To study the effect of geniposide-acid(GA) on the proliferation and inflammatory cytokines secretion of RSC-364 stimulated by culture supernatant of macrophages induced from THP-1 cells.【Methods】THP-1 cells were differentiated into macrophage induced by phorbol myristate acetate(PMA), then were stimulated by lipopolysaccharide(LPS). The supernatant fluids of macrophages were harvested. Then the synovial cells of rat RSC-364 were divided into control(medium) group, model(supernatant) group,methotrexate group(1×10^-6mol/L)and three GA groups(1×10^-5, 1×10^-6, 1×10^-7mol/L). Cell proliferation was tested by MTT assay. Cell cycle was analyzed by flow cytometry. The levels of TNF-αand IL-1βin supernatant were determined by enzyme linked immunosorbent assay(ELISA).【Results】GA could inhibit RSC-364 cells proliferations and secretions of IL-1βand TNF-αdose dependently. High dose group of GA had most significant effect compared with model group. GA at 10^-5mol/L obviously inhibited RSC-364 cells proliferations(the inhibition rate was 25%, 21% and 20% respectively at 24,48 and 72 h), increased G1-phase cell numbers of the whole cycle [(79.20+0.55)% vs(69.22+0.33)%, P〈0.01], and reduced the levels of IL-1β[(14.35±0.12) vs(40.55±0.61) ng/L, P〈0.01] and TNF-α[(11.92±0.71) vs(40.45±0.38) ng/L, P〈0.01] in supernatants of treated cells.【Conclusion】GA could significantly inhibit RSC-364 cell proliferation and inflammatory cytokines secretion of TNF-αand IL-1β, which were stimulated by supernatant of macrophage.
