摘要
目的观察筋骨痹痛丸对家兔膝骨关节炎模型软骨细胞凋亡的影响及mmp-1的表达,初步探讨筋骨痹痛丸在治疗膝骨关节炎疗效中的作用及机制。方法 36只实验性家兔随机分成正常组、模型组、筋骨痹痛丸组、氨基葡萄糖胶囊组,每组9只。除正常组外,其余3组采用石膏制动法建立膝骨关节炎动物模型:6周后,解除家兔关节的制动,筋骨痹痛丸组按7935mg/(kg·d)给予灌胃3周,氨基葡萄糖胶囊组按90mg/kg灌服3周,正常组和模型组正常饲养。9周后,取家兔膝骨关节软骨标本,原位末端标记法(TUNEL法)检测软骨细胞凋亡,免疫组化法检测mmp-1的表达。结果TUNEL法检测结果显示:筋骨痹痛丸组、氨基葡萄糖胶囊组AI明显低于模型组(P<0.05),筋骨痹痛丸组AI与氨基葡萄糖胶囊组比较无明显统计学意义;免疫组化检测结果显示:治疗后,筋骨痹痛丸组、氨基葡萄糖胶囊组mmp-1表达较模型组明显降低(P<0.05),筋骨痹痛丸组、氨基葡萄糖胶囊组比较无明显差异。结论筋骨痹痛丸能有效减轻家兔膝关节炎模型软骨的损伤,可能是通过降低软骨细胞中mmp-1的表达实现的。
Objective To observe the effect of JingGuBiTong Pill on knee joint chondrocyte apoptosis and expressions of gene mmp-1 in rabbits with knee osteoarthritis modles,investigate the mechanism of JingGuBiTong Pill treatment of knee osteoarthritis. Methods 36 rabbits were randomly divided into normal group,model group,JingGuBiTong Pill group,Glucosamine capsule group,n = 9. The rabbits in normal group,JingGuBiTong Pill group and Glucosamine capsule group were made into the models with the Knee Osteoarthritis,lift the articular brake after modeling 6 weeks. JingGuBiTong Pill group was fed with JingGuBiTong Pill 7935 mg /kg 3 weeks,group D was fed with Glucosamine capsule 90 mg /kg 3 weeks,the other groups were normal fed. After nine weeks,take the rabbit knee joint cartilage specimens,detected chondrocyte apoptosis by TUNEL,detected the expression of mmp-1 by immunohistochemical method. Results TUNEL showed that: AI of JingGuBiTong Pill group and Glucosamine capsule group was significantly lower than the model group(P〈0. 05),the difference between JingGuBiTong Pill group and Glucosamine capsule group was no statistically significant. Immunohistochemistry showed: After treatment,mmp-1 expression of JingGuBiTong pill group and glucosamine capsule group was significantly lower than the model group(P〈0. 05),JingGuBiTong pill group,glucosamine capsules group showed no significant difference. Conclusion JingGuBiTong pills can effectively reduce the damage of knee joint chondrocyte in rabbits with knee osteoarthritis models,probably by reducing the mmp-1 expression of knee joint chondrocyte.
