摘要
目的研究苗药"四大血"对佐剂性关节炎大鼠的抗炎作用,并探讨其作用机制。方法 60只大鼠随机分成空白组、模型对照组、"四大血"高、中、低剂量组和阳性对照组,每组10只。除空白组外,其余各组采用足趾部皮下注射弗氏完全佐剂,建立佐剂性关节炎大鼠(AA)动物模型。观察各组大鼠右足跖外观变化、肿胀程度、滑膜组织病理特征;Elisa法检测大鼠膝关节滑膜组织炎性细胞因子白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)表达水平。结果给药后,"四大血"中、高剂量组及阳性对照组足跖肿胀程度明显小于模型对照组(P<0.05);病理切片显示,"四大血"中、高剂量组较模型对照组大鼠滑膜组织充血减轻,炎性细胞浸润减少。酶联免疫吸附试验(Elisa)检测显示与模型对照组比较,"四大血"能下调IL-1β和TNF-α的表达量(P<0.05,P<0.01)。结论 "四大血"有抑制AA大鼠滑膜组织炎症的作用,减少滑膜组织炎性细胞浸润及滑膜上皮细胞增生,其作用机制可能与下调炎症介质IL-1β、TNF-α的表达有关。
Objective To investigate the effects of traditional Miao drug ＂Sidaxue＂on Rheumatoid Arthritis（ RA） rats and to explore its therapeutic mechanism. Methods 60 rats were randomized into six groups： blank group,model group,＂Sidaxue＂treatment high dose group,＂Sidaxue＂treatment middle dose group,＂Sidaxue＂treatment low dose group,positive control group. Except the blank group,rats in other groups were given paw subcutaneous injection of Complete Freund＇s Adjuvant（ CFA） to establish the model of RA; observed the paw changes in appearance,joint swelling degree,pathological changes of the synovial cells. Elisa was adopted to measure the expression level of IL- 1β、TNF- α. Results After continuous administration,compared with the model group,＂Sidaxue＂treatment high dose group,＂Sidaxue＂treatment middle dose group and the positive group had significant difference in swelling degree（ P 0. 05）; the results of HE staining showed that synovial congestion and inflammatory cell infiltration in ＂Sidaxue＂treatment middle dose group and＂Sidaxue＂treatment high dose group were relieved as compared with the model group. The inflammatory factors expression of IL- 1β and TNF- αin arthritis rats＇ synoviocyte were decreased（ P 0. 05,P 0. 01） compared with the model group. Conclusion＂Sidaxue＂has an anti- inflammaton effect by relieving joint swelling and inflammatory cell infiltration in the synovial tissue,and its therapeutic mechanism may be related to inhibiting the expression of inflammatory factors IL- 1β and TNF- α.
