摘要
采用高效液相(HPSEC-RID)体积排阻色谱法,建立了7种常见单糖的分离模式,成功地用于黄秋葵多糖的单糖组成分析。应用体积排阻色谱法,色谱柱为Polyspher CH PB(7.8 mm×300 mm,made in Germany),流动相为去离子水,柱温80℃,示差折光检测器(检测器温度40℃),流速0.6 ml/min内标为丙三醇。根据建立的方法测定,黄秋葵多糖由木糖、甘露糖、鼠李糖、葡萄糖、果糖、半乳糖及阿拉伯糖7种单糖组成,其摩尔比分别为0.57∶1.00∶0.53∶3.50∶1.75∶1.25∶1.70。该HPSEC-RID方法是简单、快速、灵敏、方便的分析技术,具有良好的重复性,可用于黄秋葵多糖的单糖组成的质量控制。
A method of analysis the monosaccharide composition of Okre polysaccharides with good reproducibility is established in thhi paper. The method is to base on HPSEC-RID method and chromatographic conditions that inc1 ude Polyspher CH PB( 7. 8 mm × 300 mm,made in Germany) column. The mobile phase is pure water at 40℃ of the column and the RID detector is 80℃. The method of HPSEC-RID analysis showed that Okra polysaccharides consists of xylose,mannose,rhamnose,glucose,fructose,galactose and arabonose with the molar ratios of 0. 57 ∶ 1. 00 ∶ 0. 53 ∶ 3. 50 ∶1. 75 ∶ 1. 25∶ 1. 70. It was proved that the HPSEC-RID method is simple,rapid,sensitive,convenient,and can be applied to the quality control of the monosaccharide composition of Okra polysaccharides.
