摘要
为建立谷子茎尖组织中植物激素的色谱分析方法,本试验采用高效液相色谱法对谷子中赤霉素(Gibberellin,GA3)、脱落酸(Abscisic acid,ABA)、α-萘乙酸(Naphthaleneacetic acid,NAA)、6-苄氨基嘌呤(6-Benzylaminopurine,6-BA)4种植物激素进行分离和测定,试样经液氮研磨后用80%甲醇浸提植物激素,提取液蒸发至无甲醇,石油醚脱色素,乙酸乙酯萃取植物激素,以Hypersil C18色谱柱分离,甲醇(A)-0.75%冰醋酸溶液(B)梯度洗脱,检测波长254nm,流速为0.6mL/min,进样体积为20μL,柱温30℃。在本试验条件下,赤霉素、脱落酸、α-萘乙酸、6-苄氨基嘌呤能与其它物质实现有效分离。4种待测物质线性关系良好,相关系数为0.998 9~0.999 9,GA3、ABA、NAA和6-BA最低检出限分别为22.4,0.1,5.1和0.2mg/kg,回收率范围为77.4%~108.8%,相对标准差小于10%。该方法选择性高、回收率稳定、重复性良好,可以用于谷子叶尖中植物激素的检测。
In order to make clear the levels of plant hormones in foxtail millet leaf tissues ,the detection method of high performance liquid chromatography (HPLC) was constructed .The leaf samples were firstly ground in liquid nitrogen ,and the plant hormones were digested with 80% methanol .Afterward ,the extracted solution was evaporated until to methanol‐free ,fol‐lowed by depigmentation with petroleum ether and then extraction of plant hormones with ethyl acetate .For the method ,the conditions were as the follows :separation with Hypersil C18 chromatographic column ,gradient elution with methanol(A) and 0 .75% ice acetic acid so‐lution(B) ,measurement wavelength of 254 nm ,flow rate of 0 .6 mL/min ,injection volume 20μL ,column temperature 30 ℃ .Results showed that ,under the condition of this test ,gibber‐ellins(GA3 ) ,abscisic acid(ABA) ,α naphthalene‐acetic acid(NAA) and 6 benzyl amino‐pu‐rine(6 BA) could be effectively separated out from other materials .The above four plant grow th regulators showed a good linear relationship with a coefficient of 0 .998 9 to 0 .999 9 . The limits of detection for GA3 ,ABA ,NAA and 6 BA were 22 .4 ,0 .1 ,5 .1 and 0 .2 mg/kg respectively ,and their percent recoveries were between 77 .4% and 108 .8% ,and their relative standard deviation was lower than 10% .Due to the characters of high selectivity ,stable per‐cent recovery and good repetitiveness ,this method can be used for plant hormone detection of foxtail millet leaf apex .
