摘要
植物激素在植物的胚胎发育、种子萌发、生长发育、开花结果等生理过程中相互调节对生物合成起着至关重要的作用.针对植物激素在植物体内含量极低且不稳定的特征,采用高效液相色谱与质谱(HPLC-MS/MS)联用的方法检测拟南芥(Arabidopsis thaliana)不同部位中的生长素(IAA)、赤霉素(GA3和GA4)、脱落酸(ABA)、茉莉酸(JA)和水杨酸(SA)6种激素的含量,同时对玉米(Zea mays)内源激素合成突变体及野生型材料的内源GA3和GA4含量进行了定量的研究.通过不断实验,优化了植物材料的前期处理方法及提取技术,提高了植物内源激素的提取和检测效率,形成了一套较为完整高效的植物内源激素检测方法.
The regulation of plant hormones in the physiological processes such as embryo development,seed germination,growth and development,flowering and fruit are crucial to biosynthesis.Due to the low concentrations of phytohormones and unstability characteristics,high performance liquid chromatography combined with tandem mass spectrometry(HPLC-MS/MS)were used to detect the content of auxin(IAA),gibberellin(GA3 and GA4),abscisic acid(ABA),jasmonic acid(JA)and salicylic acid(SA)in different tissuses of Arabidopsis thaliana.Meanwhile,the contents of endogenous GA3 and GA4 of Zea mays mutants and wild-type materials were quantitatively studied.After continuous experiments,the plant treatment method and extraction technology were optimized,and the extraction and detection efficiency of plant hormones were improved,forming a set of relatively complete and efficient methods for the detection of plant endogenous hormones.
