摘要
目的:建立简单快速的枸杞总黄酮测定方法,同时对其水煎煮的提取动力学过程进行模拟,建立动力学方程并验证。方法:采用紫外分光光度法,对其方法学进行考察;使用该方法测定不同提取时间和溶剂倍量下枸杞水提液中总黄酮浓度,以简化的数学动力学模型为基础,推算出方程参数进行方程拟合。结果:芦丁在0.004 828-0.028 968 g·L^-1具有良好的线性关系,回归方程为Y=29.823x+0.015 8,r=0.999 6,平均回收率为99.93%,RSD=0.76%;拟合的动力学模型结果为:CB=(4.874 8×t^0.5/M-1.018 3)^0.666 256,验证结果良好。结论:所简化的枸杞总黄酮测定方法稳定可行,重现性好。模型适用并能良好的描述枸杞水提取的动力学过程。
Objective: To establish the simple and fast determination method of total flavonoids in Lycium barbarum L and study its water extraction process and deduce the kinetic model and test its accuracy. Methods: Ultraviolet Spectrophotometry was used to inspect its methodology. And the method was used to determine the concentration of total flavonoids in different extraction time( t) and solvent load( M). Based on the simplified model,correlation parameters were deduced and the equation was set up. Results: Rutin had good linear relation in the range of 0. 004 828 - 0. 028 968 g·L^- 1 and the average recovery was 99. 93% and RSD was 0. 76%. Its kinetic model was: CB=( 4. 874 8 × t^0. 5/ M- 1.018 3)^0. 666 256 and the acurancy was tested. Conclusion: The method is simple,quick and stabile and reproducible and the kinetic model can describe the extraction process of Lycium barbarum L.
