摘要
目的本研究拟初步探讨let-7f与大面积脑梗死的诊断及发展的相关性。方法采用前瞻性队列研究方法,收集神经重症ICU的缺血性脑卒中患者血标本(128例),并收集年龄、性别匹配的正常人群、脑出血患者作为对照。将血清标本进行miRNA提取、反转录及实时荧光定量PCR,以检测血清中let-7f的相对表达量。结果血清let-7f在大面积前循环脑梗死患者中表达较正常人群显著下调(P<0.05);在后循环脑梗死患者中的表达水平较正常人群无显著性差异。血清let-7f在大面积脑梗死合并出血转化患者中表达较正常人群显著上调(P<0.05),但较脑出血患者无显著性差异。血清let-7f在脑出血患者中的表达水平较正常人群显著上调(P<0.05)。血清let-7f在非大面积脑梗死患者中的表达水平较正常人群显著上调(P<0.05),但低于脑出血患者。血清let-7f表达水平与肺部感染无显著性差异(P>0.05)。血清let-7f与脑出血或脑梗死合并出血呈高度正相关性,其次与梗死面积呈负相关。结论血清let-7f表达水平对于大面积前循环脑梗死的诊断及发展具有一定的相关性。
Objective To investigate the correlation between the expression of let‐7f in serum and the diagnosis and the de‐velopment of large area cerebral infarction.Methods Prospective cohort study was used to collect the ischemic stroke patients blood samples (128 cases) in neurological ICU ,and the serum of normal people and cerebral hemorrhage patients whose age and sex were matched.The relative expression of let‐7f in plasma was checked by managing the serum samples by miRNA ex‐traction ,reverse transcription and quantitative real‐time PCR.Results The expression of let‐7f in patients of pre‐cycle massive cerebral infarction was significantly lower than normal population (P0.05).The expression of let‐7f in serum had a highly positive correlation compared with hemorrhagic infarction and encephalorrhagia ,and was negatively correlated with the infarct area.Conclusion The expression of let‐7f in serum is meaning for the diagnosis and the development of large area cerebral infarction.
