摘要
目的:探讨复方大七气汤(CDQD)对裸鼠卵巢癌细胞皮下移植瘤凋亡的促进作用,为临床治疗提供理论依据。方法:建立裸鼠卵巢癌皮下移植瘤模型,将成功造模的30只荷瘤鼠分为5组(n=6),阴性对照组、复方红豆杉胶囊组、CDQD高剂量组、CDQD中剂量组、CDQD低剂量组,给药21 d后处死所有荷瘤鼠,测瘤体积、瘤质量并绘制肿瘤生长曲线、计算抑瘤率,检测血清糖类抗原CA125,以免疫组化SP法和图像分析技术检测Survivin蛋白、Caspase-3蛋白、Caspase-8蛋白的表达。结果:CDQD汤高剂量组的血清CA125、瘤质量、癌组织Survivin的表达明显低于阴性对照组(P<0.05),抑瘤率、Caspase-3、Caspase-8蛋白的表达明显高于阴性对照组(P<0.05),与复方红豆杉胶囊组比较无明显差异。结论:CDQD对卵巢癌皮下移植瘤增殖具有抑制作用,其机制可能与促进肿瘤细胞凋亡有关。
OBJECTIVE To investigate the stimulation function that the compound da qiqi decoetion(CDQD) plays on the apoptosis of subcutaneous transplanted ovarian cancer in nude mice and to provide theory evidence for clinical treatment. METHODS Making the models of subcutaneous transplanted ovarian cancer in nude mice, and divide the 30 mice with tumor into 5 groups(n = 6), the negative control group, Compound Taxus capsules group, CDQD high dosed group, CDQD middle dosed group, CDQD low dosed group. Killing all the mice after 21days's treatment and measure the volume and weight of the tumor and calculate damps the lump rate, tumor growth curve. Test the serum levels of CA125. Examining the expression of the Survivin protein,Caspase-3 protein and Caspase-8 protein by immunohistochemical SP method. All groups were measured and compared. RESULTS The expression in serum CA125, the weight of tumor and cancer tissue Survivin protein of the CDQD with high dose is obviously weaker than that of the negative control group (P〈0. 05) ; the expression of damps the lump rate and Caspase-3 protein,Caspase-8 protein is obviously stronger than that of the negative control group (P〈0. 05) and almost equals to that of the group of the compound Taxus capsule. CONCLUSION The CDQD can inhibit the proliferation on subcutaneous transplanted ovarian cancer, whose process may be related with the stimulation on the apoptosis of tumor cells.
