摘要
左金丸组分代谢研究主要在I相和II相反应代谢物的鉴定上,但未见左金丸组分黄连碱及吴茱萸碱代谢报道。关于代谢途径方面,仅有对小檗碱的报道,其他生物碱的代谢途径未见报道。对CYP450酶影响方面,黄连对CYP1A2、CYP3A4、CYP2C9、CYP2C19、CYP2D6和CYP2E1有抑制作用,吴茱萸对CYP1A2、CYP3A4、CYP2C19、CYP2D6和CYP2E1有抑制作用,小檗碱对CYP1A2、CYP3A4、CYP2C9、CYP2D6和CYP2E1有抑制作用,黄连碱对CYP1A2和CYP2D6有抑制作用,巴马汀对CYP2D6和CYP2E1有抑制作用,药根碱对CYP3A4、CYP2C9、CYP2D6和CYP2E1有抑制作用,吴茱萸碱对CYP1A2抑制作用,吴茱萸次碱对CYP3A4、CYP2C9、CYP2C19、CYP2D6和CYP2E1有抑制作用。但也有研究发现小檗碱对CYP1A2活性没有影响,吴茱萸次碱对CYP1A2有诱导作用,小檗碱和巴马汀对CYP3A4有呈浓度依赖的激动作用,黄连碱对CYP2C9有激动作用,巴马汀对CYP2C9和CYP2C19也有激动作用。
Research of constituents Metabolism and effect for CYP 450 of Zuojin Pill is mainly on phase I and II metabolites,but Cop-tisine and Evodiamine havn't be reported. About metabolic pathway,only Berberine has been reported. Huanglian has inhibitory action for CYP1A2,CYP3A4,CYP2C9,CYP2C19,CYP2D6 and CYP2E1. Wuzhuyu has inhibitory action for CYP1A2,CYP3A4,CYP2C19, CYP2D6 and CYP2E1. Berberine has inhibitory action for CYP1A2,CYP3A4,CYP2C19,CYP2D6 and CYP2E1. Palmatine has inhibito-ry action for CYP2D6 and CYP2E1. Jatrorrhizin has inhibitory action for CYP3A4,CYP2C9,CYP2D6 and CYP2E1. Wuzhuyu has inhib-itory action for CYP1A2. Rutaecarpine has inhibitory action for CYP3A4,CYP2C9,CYP2C19,CYP2D6 and CYP2E1. But studies have found that Berberine has no effect on CYP1A2 activity,Rutaecarpine has induced effect on CYP1A2. Berberine and Palmatine have con-centration dependent activation for CYP3A4. Coptisine has activation for CYP2C9. Palmatine has activation for CYP2C9 and CYP2C19.
