摘要
目的:观察桂枝及配伍对CIA大鼠细胞因子的影响,探讨其作用机理。方法:雄性Wistar大鼠80只,随机取10只大鼠作为正常对照,其余采用Ⅱ型胶原(CⅡ)诱导的关节炎并随机分为模型、水煎桂枝、醇提桂枝、水煎肉桂、水煎桂枝附子及醇提桂枝附子组,致炎后21天开始ig给药,连续21天,观察大鼠关节炎指数并测定大鼠血清中TNF-α、IL-1β、IL-6含量。结果:水煎桂枝、桂枝附子及醇提桂枝附子组关节炎指数明显减轻(P<0.05或P<0.01);与模型组比较,水煎桂枝、肉桂、桂枝附子及醇提桂枝附子组TNF-α明显降低(P<0.05),且水煎桂枝附子组对IL-6下调作用明显(P<0.05)。结论:桂枝及桂枝附子配伍对CIA大鼠血清TNF-α和IL-6有不同的下调作用,这可能是其治疗RA作用机理之一。
Objective:To observe the Cassia twig and compatibility influence on the concentration of cytokines in the CIA rats , explore the action mechanism of Cassia twig compatibility on rheumatoid arthritis. Methods : 1 0 rats were randomlyselected from 80 male Wistar rats as control group, the left 70 rats were adopted type 11 collagen - induced autoimmunear- thritis. Autoimmune arthritis rats were randomly divided into model group, cassia twig of water decoction group, cassia twig of alcohol extract group, cinnamon of water decoction group, cassia twig Monkshood of water decoction group, cassia twig Monkshood of alcohol extract group and started to be intragastric administration last for 21 days. TNF - et, IL - 113 and IL - 6 activity in the CIA rat model were deected by the Enzyme - linked immunoadsordent assay (Elisa) and the ar- thritis index were observed after intervention. Results : Compared with that of model group, Cassia twig of water decoction group , Cassia twig Monkshood of alcohol extract group, Cassia twig Monkshood of water decoction group arthritis index were reduced significantly ( P 〈 0.05 or P 〈 0.01 ). The activity of TNF - a, IL - 1β and IL - 6 in model group were sig- nificant higher than the normal group. Compared with the control group, Cassia twig of water decoction group, Cinnamon of water decoction group, Cassia twig Monkshood of water decoction group and Cassia twig Monkshood of alcohol extract group significantly reduces TNF - a( P 〈 0.05 ) ; and Cassia twig Monkshood of water decoction group significantly reduces IL - 6 ( P 〈 0. 05 ). Conclusion : The Cassia twig and Cassia twig with Monkshood compatibility can decrease TNF - a, IL -6 which rise abnormally in serum, this may be one of mechanisms which cure RA.
