摘要
采用实时荧光定量PCR技术,以GAPDH为内参,检测鹿苑鸡在胚胎期9、11、13、15、17、19、21日胚龄时肝脏中CROT、HADHB、SREBP2 3个基因的表达量变化。结果显示:肝脏中CROT表达量最高的是21日胚龄(E21)胚胎,且其表达量随着日胚龄的增加先上升后下降然后又上升,其中2个峰值所对应的是E13和E21胚胎,CROT表达量最低的是E17胚胎;HADHB基因表达与CROT相似,表达量最高的是E21胚胎,最低的是E17胚胎,表达量随着日胚龄的增加先上升后下降然后又上升;SREBP2基因在肝脏中表达量最高的也是E21胚胎,与其他日胚龄胚胎的表达量差异极显著。结果揭示了鹿苑鸡胚胎肝脏中CROT、HADHB、SREBP2 3个基因m RNA的表达特征,为从分子水平了解鸡脂类代谢的调控规律奠定理论基础。
The real-time fluorescence quantitative PCR was applied to investigate the tissue expression patterns of CROT, HADHB and SREBP2 mRNA in liver of Luyuan chicken at 9, 11,13,15, 17, 19 and 21 days of embryonic stage （E9,E11,E13,E15,E17,E19 and E21）,and GAPDH was used as inner control. The results showed that the highest expression level of CROT gene in liver was at E21, the expression of which increased with the days of embryonic stage increasing at first, then decreased and increased with the development of embryo, the two corresponding peaks were at El3 and E21, the lowest expression level of CROT in liver was at El7. The expression pattern of HADHB gene was similar to CROT, the highest expression level of HADHB in liver was at E21,the lowest expression of HADHB in liver was at E17,its expression level increased with development at first,then decreased and increased with development of embryo. The highest expression level of SREBP2 in liver was also at E21, it was significantly different from other embryonic ages. The experiment results revealed the expression characteristics of CROT,HADHB and SREBP2 mRNA in the liver of chicken embryo,and laid the theoretical foundation for understanding the regulation of chicken lipid metabolism at the molecular level.
