摘要
目的探讨微小RNA-145(miR-145)对结肠癌细胞HCT116增殖、侵袭和迁移能力的影响。方法采用慢病毒转染将过表达miR-145质粒(A组)和空白质粒(B组)分别转入HCT116细胞,通过CCK-8实验、Transwell侵袭实验和划痕实验分别检测两组细胞增殖、侵袭和迁移能力。结果细胞转染后第3、4和5天,A组细胞增殖能力较B组降低(0.6612±0.0015vs.1.0194±0.0024、1.1680±0.0002vs.1.7186±0.0009和1.2530±0.0006vs.2.2261±0.0064)(P<0.05)。A组平均穿膜细胞数和细胞迁移数低于B组[(122±18)个vs.(240±26)个和(156±28)个vs.(266±26)个](P<0.05)。结论 miR-145能降低HCT116细胞的增殖、侵袭和迁移能力。
Objective To explore the effects of microRNA-145(miR-145) on the proliferation, invasion and migration of colon cancer cell line HCTll6. Methods Plasm{ds of miR-145(group A) and empty plasmid (group B) were separately transfected into HCT116 by lentivirus. The proliferation, invasion and migration of HCT116 cells were tested by CCK-8 assay, transwell assay and wound healing assay, respectively. Results The proliferation of HCT116 cells in group A was lower than that in group B on the 3rd ,4th and 5th day after cell transfection(0. 6612±0. 0015 vs. 1. 0194± 0.0024,1.1680±0.0002 vs. 1.7186±0.0009 and 1. 2530±0.0006 vs. 2.2261±0.0064) (P〈0.05). Compared with group B, the invasion and migration o{ HCTll6 cells were reduced in group A[( 122 ± 18) pieces vs. (240 ± 26) pieces and ( 156 ± 28) pieces vs. ( 266 ± 26) pieces-] (P〈0. 05). Conclusion miR-145 can inhibit the proliferation, invasion and migration of HCT116 cells.
