Expression of microRNA-145 in non-small cell lung cancer tissues and its correlation with prognosis

摘要 目的 探讨微小RNA-145在非小细胞肺癌组织的表达及其与预后的关系.方法 分析江苏省肿瘤医院2005年11月至2007年1月行手术治疗的104例非小细胞肺癌患者的临床资料.采用实时定量逆转录聚合酶链反应(RT-qPCR)检测上述病例肺癌标本的微小RNA-145水平;应用x2,检验分析微小RNA-145表达水平与临床病理参数的关系;采用Kaplan-Meier方法分析微小RNA-145的表达与非小细胞肺癌患者生存期之间的关系;采用Cox风险比例模型分析各个协变量的联合效应.结果 微小RNA-145的表达量与患者年龄(x2=0.155,P>0.05)、性别(x2=1.251,P>0.05)、病理类型(x2=0.452,P>0.05)、术后外科病理分期(p-TNM,x2 =0.156,P>0.05)及吸烟史(x2 =2.6,P>0.05)无明显相关;生存分析显示微小RNA-145低表达组较高表达组患者生存期明显缩短(P<0.01).Cox风险比例模型分析显示微小RNA-145低表达可作为非小细胞肺癌患者预后不良的标志.结论 微小RNA-145低表达提示非小细胞肺癌患者预后较差. Objective To investigate the expression of microRNA (miR)-145 in non-small-cell lung carcinoma (NSCLC) tissues and its correlation with prognosis.Methods The clinical features of 104 NSCLC patients who underwent surgery in our hospital from November 2005 to January 2007 were analyzed.Real-time quantitative polymerase chain reaction (RT-qPCR) was used to detect the expression of miR-145.Chi-square test was utilized to analyze the relationship between miR-145 expression and clinicopathological features.Survival curves were plotted by using the Kaplan-Meier method and compared by the log-rank test.A Cox proportional hazard regression model was applied to examine the joint effect of covariables.Results No significant correlation was found between the expression of miR-145 and age (x2 =0.155,P 〉0.05),gender (x2 =1.251,P〉0.05),pathological type (x2 =0.452,P〉0.05),p-TNM (x2 =0.156,P 〉 0.05),or smoking (x2 =2.6,P 〉 0.05).Kaplan-Meier analysis showed that low expression of miR-145 was associated with a shorter survival time (P 〈 0.01).Finally,both univariate and multivariate analyses demonstrated that low miR-145 expression might be a poor prognostic marker of NSCLC patients.Conclusion Down-regulation of miR-145 was associated with poor prognosis of patients with NSCLC.
