制备型高效液相色谱在药物分离纯化中的应用研究进展 被引量:7

Advances in drug separation and purification by preparative high performance liquid chromatography

摘要 以纯化中药和天然产物活性成分、分离合成药物中含有的杂质及制备生物医药为重点,阐述了分离不同类型化合物采用的固定相、流动相等优化条件。药物的分离纯化多采用以C18为填料的反相色谱柱。中药及天然产物的纯化所采用的溶剂体系多为甲醇水溶液或乙腈水溶液,为调节洗脱强度可以添加H3PO4、冰乙酸、正丁烷、异丙醇等。分离合成药物中的杂质通常采用添加磷酸盐、乙酸铵的甲醇水或乙腈水溶液体系。符合制备色谱流动相应质优价廉,后处理容易的选择原则。 The application of preparative high performance liquid chromatography in separating drugs are summarized,including the purification of the active ingredients in traditional Chinese medicine and natural products,the separation of impurities in synthetic drugs and preparation of biological medicine. The stationary phase and mobile phase used to separate different types of compounds are discussed. The results showed that the reversed phase chromatography can be used in most process of separation and purification for drugs. The solvent systems for purification of traditional Chinese medicine and natural products were usually adopted the aqueous solution of methanol or acetonitrile solution with phosphoric acid,glacial acetic acid,n-butane,isopropyl alcohol,etc. to adjust elution strength. The solvent systems for the synthetic drug separation usually were adopted the above basic solution system with phosphate,ammonium acetate. These conform to the selection principles that the mobile phase should be high quality,cheap and post-processing easily.
