摘要
目的:比较不同浓度醇沉的瓜蒌薤白提取液的抗氧化性及其药效,以期为瓜蒌薤白提取液的精制选择适当体积分数的乙醇提供理论依据。方法:采用1,1-二苯基-2-苦基肼自由基(DPPH)测定法对5组(未醇沉组与95%,85%,75%,65%乙醇醇沉组)瓜蒌薤白提取液的抗氧化活性进行分析;采用小鼠腹腔注射异丙肾上腺素(Iso)造模,以模型组为对照,测定5组瓜蒌薤白提取液灌胃的小鼠心电图T波变化及血浆中一氧化氮(NO)、超氧化物歧化酶(SOD)含量,考察各组药效。结果:75%醇沉组的抗氧化活性最强,且与未醇沉组相比,有统计学意义(P<0.05);与模型组比较,75%醇沉组在5,10,15,20,30 s,1,2,5,10 min几个时间点能明显抑制T波的改变,且有统计学意义(P<0.05或0.01);与模型组比较,各组均能显著提高小鼠血浆中NO含量,且有统计学意义(P<0.01);与模型组比较,65%、75%和85%醇沉组能显著提高小鼠血浆中SOD含量,且有统计学意义(P<0.05,其中75%醇沉组P<0.01)。结论:75%醇沉组的抗氧化活性及对Iso诱导的小鼠急性心肌缺血模型的保护作用最为显著,可用75%乙醇对瓜蒌薤白提取液进行精制处理。
OBJECTIVE To compare the antioxidant activity of Gualou Xiebai extracts percipitated by different alcohol con- centrations, in order to provide theoretical basis for choosing the proper concentration in the refining of the extract. METHODS The antioxidant activities of five groups extractions (non-alcohol deposit fluid, 95 % alcohol deposit fluid, 85 % alcohol deposit fluid, 75 % alcohol deposit fluid, 65 % alcohol deposit fluid) were analyzed by DPPH assay. Mice model induced by isoproterenol was employed as control group. The five test groups were caused by intragastric administration respectively using 5 groups of extractions. The efficacy of each group was evaluated by comparing ECG T wave changes and the plasma contents of NO and SOD. RESULTS The protective effect of 75 % alcohol deposit fluid group on antioxidant activity was the strongest, it had sta- tistically significant difference compared with non-alcohol deposit fluid group (P〈0. 05) ; compared with model group, 75 % al- cohol deposit fluid group significantly inhibited the ECG T wave changes at different time point which included 5 s, 10 s, 15 s,20 s,30 s, 1 min,2 min,5 min and 10 min, (P〈0. 05 or 〈0. 01) ; all the test groups significantly increased the plasma contents of NO compared with model group (P〈0. 01) ;the 65%,75% and 85% alcohol deposit fluid groups significantly increased the plasma contents of SOD compared with model group (P〈0. 05, which 75% alcohol deposit fluid group P〈0. 01). CONCLU- SION The 75 % alcohol deposit fluid group had the most significant protective effect on myocardial ischemia mice induced by isoproterenol, the GualouXiebai extracts could be prepared by using 75 % ethanol to refine.
