摘要
目的 :观察补肾益智方对AD动物模型大脑神经元磷酸化Tau蛋白及其相关酶类蛋白磷酸酶 1(PP 1)、蛋白磷酸酶 2A(PP 2A)的影响。方法 :用Wistar大鼠 80只以D 半乳糖腹腔注射 4周加上鹅膏覃酸 (ibotenicacid ,IBO)脑内Meynert核注射制造AD模型 ,随机分成AD模型组、双益平对照组、补肾益智方高、低剂量治疗组加上正常老年组和正常青年组总共设立六个组别。动物经治疗处理 6周后 ,应用免疫组织化学方法检测大鼠海马CA1和齿状回磷酸化Tau蛋白、PP 1、PP 2A神经元数目和光密度。结果 :AD模型组海马内神经元的Tau蛋白过度磷酸化而相互集结 ,形成成对螺旋细丝 ,表达脱Tau蛋白磷酸化的磷酸酯酶PP 1、PP 2A均明显减少 ;而补肾益智方治疗组可以使这种现象得到明显改善。结论 :补肾益智方能较有效地改善AD大鼠脑内神经元的Tau蛋白过度磷酸化 ,促进PP 1、PP 2A的表达。
Objective:To observe the effects of Bu Shen Yi Zhi Decoction on phosphated Tau protein,protein phosphataes 1(PP 1) and protein phosphataes 2A(PP 2A) of hippocampal neurons of AD model rats.Method:Eighty fifteen month Wistar rats were induced by D galactose intraperitoneal injection and lesion of basal nucleus of Meynert with ibotenic acid to make AD model,randomly divided into AD model group,Hup A treated group,BSYZD (high dose and low dose) treated group,and normal control groups.After treatmenting,tissue speciments for each group were removed from the hippocampus CA1 area for light microscopy studies.Cryostat sections were studied with immunohistochemistry for phosphated Tau protein,protein phosphataes 1(PP 1),protein phosphataes 2A(PP 2A).Results:The phosphated tau protein of hippocampal neurons of AD model rats showed abnormally assembled into paired helical filaments and decrease the expression of tau dephosphorylating enzymes (PP 1 and PP 2A),while the use of Bu Shen Yi Zhi Decoction can significantly ameliorate these changes.Conclusion:The Bu Shen Yi Zhi Decoction can significantly ameliorate the phosphated tau protein and the expression of the enzymes PP 1 and PP 2A of hippocampal neurons of AD rats.
