松花粉水提取物对四氯化碳诱导肝细胞损伤的保护作用 被引量:3

摘要 目的 探讨松花粉水提取物在体外对人正常肝细胞氧化损伤的保护作用。方法 应用体系终浓度为40mmol/L四氯化碳(CCl4)条件培养液制备体外人肝细胞系L-02氧化损伤模型.同时加入不同浓度的松花粉提取物共培养,通过水溶性四氮唑(WST-1)法检测肝细胞增殖活性,全自动生化分析仪检测肝细胞上清液转氨酶活性,病理切片观察肝细胞形态变化。结果 CCl4对肝细胞增殖活性、功能及形态均有明显毒性作用,可使肝细胞增殖活性降低29.8%。38.4%,天冬氨酸氨基转移酶(AST)增高22.3%,丙氨酸氨基转移酶(ALT)增高99.2%;正常肝细胞形态发生明显变化,胞核胞浆固缩,细胞生长排列混乱。松花粉提取物干预对上述细胞的增殖活性保护作用不显著.但可使CCl4损伤肝细胞的AST降低14.8%。27.3%,ALT降低39.8%~70.1%,差异有统计学意义(P〈0.05);并对肝细胞胞浆胞核固缩现象有明显改善,高浓度松花粉(400mg/L)干预组细胞形态接近正常,ALT明显降低(P〈0.05)。结论 松花粉提取物对CCl4损伤L-02细胞的增殖活性保护作用不明显.但对肝细胞形态和功能损伤具有有效保护作用。 Objective To observe the protective effect of masson pine pollen aqueous extracts (MPPAE) on oxidative damage of human liver cell line L-02 by CCI4. Methods Oxidative damage model of human liver cell line L-02 by CC14 (with the final concentration of 40 retool/L) had been built via cell culture in vitro. The effects of different concentrations of MPPAE on cell proliferation, morphology and functional changes after the injury were observed. Results CCl4 had obvious toxicity on the proliferation activity, cell morphology and function of liver cells. It delivered 29.3%-38.4% decrease in proliferation activity and 22.3% and 99.2% increase in activity of AST and ALT. Liver ceils grew in a disordered manner and the nucleus and cytoplasm became pyknosis. The MPPAE had no obvious protective effect on the proliferation of L-02 cells. But it had obvious positive effects on the CCL induced liver cell morphology changes and inhibited the abnormal raise of ALT (39.8%-70.1%) and AST (14.8%-27.3%) with a dose effect pattern. The high dose (400 mg/L) intervention of MPPAE could ameliorate the changes of nucleus deformity to almost normal in appearance. Conclusion MPPAE has the protective effect of cell morphology and function of liver cells although it has limited protection on the proliferation of liver cells.
