摘要
目的:观察华蟾素注射液对H22肝癌小鼠肿瘤组织中Caspase-8、Caspase-3基因(mRNA)及蛋白表达的影响。方法:40只ICR小鼠采用皮下接种H22肝癌细胞株的方法复制小鼠肝癌模型,造模成功后,将小鼠随机分为4组:模型组,华蟾素注射液低、高剂量组,顺铂(DDP)组。观察各组小鼠肿瘤生长情况变化。采用荧光定量RT-PCR方法和Western Blot检测方法检测各组H22肝癌小鼠肿瘤组织中Caspase-8、Caspase-3基因及蛋白表达的影响。结果:华蟾素低剂量组不能有效抑制肿瘤的生长,而华蟾素高剂量组、DDP组的抑瘤率分别为41.67%、52.78%,具有明显的抑制肿瘤生长作用(P<0.01)。与模型组比较,华蟾素低剂量组小鼠肝癌组织中的Caspase-8、Caspase-3表达无明显差异,而华蟾素高剂量组、DDP组能促进肝癌组织中的Caspase-8、Caspase-3的mRNA的表达升高(P<0.05)及蛋白表达明显升高(P<0.01)。结论:华蟾素注射液具有抑制肝癌细胞生长的作用,其分子机制可能是上调肿瘤组织Caspase-8、Caspase-3基因及蛋白的表达,从而促进肝癌肿瘤细胞发生凋亡,达到抗肿瘤作用。
Objective: To investigate the effect of Cinobufacini Injection on Caspase- 8 and Caspase- 3 expressions of Gene( mRNA) and protein in tumor tissues of mice bearing Hepatoma H22. Methods: Forty ICR mice were randomly divided into 4 groups: the model group,the low,high dosage groups of Cinobufacini Injection,the Cisplatin group. After the mice model bearing Hepatoma H22 were established by subcutaneous inoculating tumor cells,the tumor growth was observed in mice. The mRNA and protein expressions of Caspase- 8 and Caspase- 3 in tumor tissues were detected by quantitative real- time reverse-transcription polymerase chain reaction( RT- PCR) and Western Blot. Results: The low dosage of Cinobufacini Injection could not inhibit the growth of tumor. The cancer inhibition ratio of the high dosage of Cinobufacini Injection and Cisplatin group was 41. 67% and 52. 78% respectively and it showed obvious inhibitory effect on tumor growth( P〈0. 01). Compared with the model group,the expression levels of Caspase- 8 and Caspase- 3 mRNA and protein used by the low dosage of Cinobufacini Injection had no obvious difference while the high dosage of Cinobufacini Injection and Cisplatin group could promote the expression levels of Caspase- 8 and Caspase- 3 mRNA( P〈0. 05) and protein( P〈0. 01) in mice tumor tissues of liver cancer. Conclusion: Our results indicated that the molecular mechanism of inhibition of the liver cancer growth treated by Cinobufacini Injection may be through the up- regulating the expression levels of Caspase- 8 and Caspase- 3 mRNA and protein so as to promote tumor cell apoptosis and achieve anti- tumor effect.
