摘要
目的探讨与瓜蒌、白及或半夏配伍对草乌急性毒性的影响。方法采用动物急性毒性实验,以动物死亡数为观测指标,计算草乌醇提液灌服给药的LD50;固定草乌剂量在LD50,与13个不同比例的反药瓜蒌、白及或半夏配伍,探讨草乌配伍禁忌的毒性变化;选用上述实验中死亡率为75%的配伍比例,即与瓜蒌(36∶1)配伍或白及(10.8∶1)配伍,测算草乌与反药配伍的LD50。结果草乌醇提液灌服给药的LD50为8.45 g·kg^-1,95%置信区间为7.58-9.43 g·kg^-1。与瓜蒌或白及配伍草乌的毒性随着瓜蒌或白及用量的增大而增强;而草乌与半夏配伍小鼠死亡率没有出现上升,反而随着半夏用量的增大呈现下降的趋势。与瓜蒌(36∶1)配伍草乌配伍禁忌的LD50为6.45 g·kg^-1,95%置信区间为5.64-7.34g·kg^-1;与白及(10.8∶1)配伍草乌配伍禁忌的LD50为5.94 g·kg^-1,95%置信区间为5.21-6.73 g·kg^-1。结论草乌与瓜蒌或白及配伍的急性毒性强于单独草乌的急性毒性;但草乌与半夏配伍的毒性没有上升,表明草乌与反药配伍禁忌不是绝对的。
Objective To investigate the acute toxicity of Aconiti kusnezoffii Radix with its anti-drugs Methods Animal acute toxicity method was established to calculate the LD50 of Alcohol extract of Aconiti kusnezoffii Radix,to research changes of toxicity of combination Aconiti kusnezoffii Radix(its dose also keep on LD50) with its anti-drugs in the article,regarding animal mortality as target. Results The LD50 of single Aconiti kusnezoffii Radix is 8. 45 g ·kg^-1,95% confidence interval is 7. 58 - 9. 43 g·kg^-1. According increase proportion of anti-drugs,the acute toxicity of co-decoction with Trichosanthes fructus,or bletillae rhizome have rised. But that of co-decoction with Pinelliae Rhizoma have reduced instead of rise trend; The LD50 of co-decoction with Trichosanthes fructus is 6. 45 g·kg^-1,95% confidence interval is 5. 64 - 7. 34 g·kg^-1; the LD50 of co-decoction with bletillae rhizome is 5. 94 g·kg^-1,95% confidence interval is 5. 21 - 6. 73 g·kg^-1. Conclusion It is revealed that the acute toxicity of co-decoction with Trichosanthes Fructus,and Bletillae Rhizome is more than that of single Aconiti kusnezoffii Radix decoction while that of co-decoction with Pinelliae Rhizoma has reduced. It is not absolutely that incompatibility of Aconiti kusnezoffii Radix with its anti-drugs.
